三氮脒处理后犬吉氏巴贝斯虫转录组测序分析

摘  要:采用转录组学测序的方法筛选三氮脒药物处理后犬吉氏巴贝斯虫差异表达基因。采集感染犬吉氏巴贝斯虫病犬的血液,用终浓度为800 nmol/L的三氮脒处理24 h,提取RNA进行转录组测序,比较三氮脒处理后犬吉氏巴贝斯虫基因表达的差异。对测序结果进行无参转录组分析,经筛选共获得77个巴贝斯虫属的基因,将筛选后的基因进行GO和KEGG数据库分析,其注释富集基因最多的类别是细胞组分、分子功能和生物进程,在KEGG通路注释中,显示与翻译通路有关的基因较多。对三氮脒处理组与未处理组的样品进行组间差异基因表达分析,发现基因CL118转录水平显著性下调。采用real-time PCR方法进行验证,结果显示本实验室保存虫株以及临床野生虫株经三氮脒处理24 h后,CL118基因的转录水平均发生下调。结果表明,基因CL118可能是三氮脒药物作用于犬吉氏巴贝斯虫虫体的潜在作用靶点。The differentially expressed genes of Babesia gibsoni(B.gibsoni) treated with diminazene aceturate were screened using the method of transcriptomics sequencing.The blood was collected from the dog infected with B.gibsoni and then treated with diminazene aceturate(800 nmol/L) for 24 h.The RNA was extracted from the lysed erythrocytes for transcriptomics sequencing.Altered transcription levels of B.gibsoni genes were confirmed based on the transcriptomics data.The sequencing results were analyzed by nonparametric transcriptome analysis.After filtering, a total of 77 genes related to Babesia were obtained.The gene ontology(GO) terms and Kyoto Encyclopedia of Genes and Genomes(KEGG) pathway analysis were applied to identify the key genes and pathways involved in the mechanism of eliminating B.gibsoni by diminazene aceturate.The results showed that the most annotated-enriched genes were involved in cell components, molecular functions, and biological processes.For the KEGG pathway annotation, the most genes were related to translation.Comparing to control group, the CL118 gene was significantly down-regulated in diminazene aceturate treatment group.The real-time PCR results also showed that the CL118 gene expression was down-regulated after treatment with diminazene aceturate in laboratorial isolates and clinical wild isolates, which was in line with the results of transcriptome sequencing.The above results indicate that CL118 gene is related to the metabolism of diminazene aceturate, and may be a potential drug target of B.gibsoni.
