摘 要:为了研究血氨对奶牛胚胎附植和妊娠的影响,本试验使用氯化氨处理奶牛子宫内膜上皮细胞,CCK8检测氨离子对细胞活力的影响,实时荧光定量RT-PCR和Western blot检测凋亡相关基因(Bax、Bcl-2、p53、Caspase-9、Caspase-8、Caspase-3)、容受性相关基因(VEGF、LIF、EGF)的mRNA和蛋白表达的变化。结果表明,5,10,15,25 mmol/L的氯化氨可显著降低细胞活力(P<0.05);经过5 mmol/L的氯化氨处理,显著增加了Bax、caspase-3、caspase-8和caspase-9的mRNA和蛋白的表达水平(P<0.05),显著下调了LIF、VEGF和EGF的mRNA和蛋白表达水平(P<0.05)。说明高浓度的血氨会诱导子宫内膜上皮细胞的凋亡和子宫容受性的降低,影响奶牛的繁殖力。In order to investigate the effects of blood ammonia on embryo implantation and pregnancy of dairy cows,endometrial epithelial cells were treated with ammonia chloride and influence of ammonia ions on cell viability was detected by CCK8 assay.RT-PCR and Western blot were used to detect the change of mRNA and protein of apoptosis-related genes(Bax,Bcl-2,p53,caspase-9,caspase-8,caspase-3)and receptivity-related genes(VEGF,LIF,EGF).The results showed that ammonia chloride of 5,10,15 and 25 mmol/L significantly decreased cell viability(P<0.05).The mRNA and protein expression of Bax,caspase-3,caspase-8 and caspase-9 were significantly increased(P<0.05),and the mRNA and protein expression of LIF,VEGF and EGF were significantly decreased(P<0.05)after 5 mmol/L of ammonia chloride treatment.These results indicated that high concentration of blood ammonia induced the apoptosis of endometrial epithelial cells and decreased the receptivity of uterus and further influenced the fecundity of dairy cows.
