摘 要:目的研究氧化苦参碱(OMT)对高交感活性诱导大鼠实验性心室肥厚的改善作用。方法SD大鼠随机分为对照组、模型组、OMT高、低剂量组、普萘洛尔组,各组于造模前1 w开始灌胃给药,除对照组外,其余各组均连续腹腔注射去甲肾上腺素(NE)15 d复制实验性心肌损伤模型,造模结束后检测大鼠血压指标、取材检测心室重构参数及羟脯胺酸含量,qRT-PCR法检测Notch-1信号变化。结果OMT能够改善大鼠血压变化,降低心室重构参数及羟脯胺酸含量,改善Notch-1信号变化。结论OMT对高交感活性诱导的实验性心肌肥厚有显著的改善作用,可能与降低Notch-1信号的mRNA表达有关。Objective To investigate the ameliorated effect of oxymatrine(OMT)on experimental ventricular hypertrophy induced by high sympathetic activity in rats.Methods SD rats were divided into 5 groups as following:control group,model group,OMT high dose and low dose groups,propranolol group.Rats in each group were given the drug by intragastric administration 1 week before model establishment.The rat model of experimental myocardial hypertrophy was established by exposed to norepinephrine(NE)by intraperitoneal injection for 15 d in all groups except the control group.At the end of modeling,the blood pressure indexes,ventricular remodeling parameters and myocardial hydroxyproline content were determined after remodeling.Meanwhile,the Notch-1 signal was detected by qRT-PCR.Results OMT could improve blood pressure,reduce ventricular remodeling parameters and the content of hydroxyproline in myocardium of rats,improve Notch-1 signal changes.Conclusions OMT could significantly improve experimental myocardial hypertrophy induced by sympathetic overactivity,which is related to the reduction of Notch-1 signal mRNA expression.
