摘 要:为了研究非洲猪瘟(ASF)暴发后我国猪繁殖与呼吸综合征病毒(PRRSV)的流行变化,本研究经PCR检测了2017年1月~2020年12月全国18个省、市和自治区1374份疑似PRRSV感染的病料样品,并对其中阳性病料样品的ORF5、nsp2基因测序并统计阳性样品的检出率,分析不同地区在ASF暴发前后PRRSV阳性检出率的变化趋势;分别利用GraphPad Prism 8.0及ORF5基因的进化树分析ASF暴发前后我国各亚型PPRSV及各地区PPRSV的流行变化趋势;根据nsp2基因推导氨基酸序列分析其缺失特征。ORF5基因和nsp2基因测序及统计分析结果显示,共检出PRRSV阳性病料样品779份(779/1374),其中ASF暴发前397份,ASF暴发后382份。ASF暴发后(2019年~2020年)我国各地PRRSV阳性检出率均有上升,华北地区PRRSV阳性检出率由60.47%上升至66.33%;东北地区由52.76%上升至53.27%;华东地区由60.93%上升至62.59%;中南地区由53.43%上升至57.50%。ASF暴发前后我国各亚型及各地区PPRSV流行变化趋势统计分析结果显示,ASF暴发后,NADC30-like PRRSV作为主要流行株,其检出率显著增长,从47.36%上升至65.97%;NADC34-like PRRSV的检出率也明显增长,从1.76%上升至5.50%;但HP-PRRSV、VR2332-like PRRSV和QYYZ-like PRRSV的检出率明显降低,分别从34.01%降至20.68%、6.80%降至3.40%、7.30%降至0.52%。可见ASF暴发后,我国大部分地区(华北、东北、华东、中南)NADC30-like PRRSV的流行趋势有所上升,但HP-PRRSV的流行趋势明显下降,并且随着ASF的流行,除中南地区外,我国各地NADC34-like PRRSV检出率明显上升。ORF5基因的遗传演化分析结果显示,ASF暴发前(2017年~2018年)检测到6种基因亚型的PRRSV(NADC30-like PRRSV、HP-PRRSV、NADC34-like PRRSV、VR2332-like PRRSV、QYYZ-like PRRSV、Type I PRRSV),而ASF暴发后还检测到CH-1a like PRRSV。Nsp2氨基酸缺失特征分析结果显示,ASF暴发前后我国各亚型PRRSV Nsp2氨基酸序列特征均未发生变化。以上结果表明,ASF疫情的暴In order to study the epidemiological changes of reproductive and respiratory syndrome virus(PRRSV)in China after the outbreak of African swine fever(ASF),1374 samples that suspected PRRSV infection from 18 provinces,cities and autonomous regions of China from January 2017 to December 2020 were detected by PCR.The ORF5 genes and nsp2 genes of the positive samples were sequenced.The detection rate of PRRSV positive samples and that of change trend before and after ASF outbreak in different regions were analyzed,and the epidemic trend of PRRSV in various subtypes and regions of China before and after the outbreak of ASF was studied via Graphpad prism 8.0 and evolutionary trees based on ORF5 genes.As well as,the amino acid sequences were deduced to confirm the deletion characteristics of the nsp2 genes.The results showed that 779 samples(779/1372)are positive for PRRSV infection including 397 positive samples before ASF outbreak and 382 positive samples after that.The result of statistical analysis confirmed that the positive rate of PRRSV is increased in all detected regions of China after the outbreak of ASF(2019-2020).The positive rate of PRRSV increased from 60.47%to 66.33%in North China,from 52.76%to53.27%in Northeast China,from 60.93%to 62.59%in East China,and from 53.43%to 57.50%in Central South China.The statistical analysis of the epidemic trend of PRRSV in various subtypes and regions in China showed that NADC30-like PRRSV is the main epidemic strain in China,its detection rate increase significantly from 47.36%to 65.97%,and the detection rate of NADC34-like PRRSV increase from 1.76%to 5.50%.While the detection rate of HP-PRRSV,VR2332-like PRRSV and QYYZ-like PRRSV decreased significantly from 34.01%to 20.68%,6.80%to 3.40%,7.30%to 0.52%after ASF outbreak.These results suggested that the epidemic trend of NADC30-like PRRSV increase in various regions of China(North China,Northeast China,East China and Central South China)after the outbreak of ASF,however,that of HP-PRRSV is reduced.In addition,the detection
