摘 要:为评价猪圆环病毒2型(PCV2)病毒样颗粒(VLP)和塞内卡病毒(SVV)全病毒灭活二联疫苗免疫效果,本研究将PCV2-VLP与SVV全病毒灭活疫苗等体积混合,加入ISA201佐剂乳化制备成PCV2-VLP和SVV全病毒灭活二联疫苗,并以PCV2-VLP疫苗和SVV全病毒灭活疫苗分别作为对照,3组疫苗免疫仔猪后于不同时间采集血清,通过间接ELISA检测血清中PCV2特异性抗体,采用中和试验法检测SVV中和抗体,结果显示单独使用PCV2疫苗免疫组仔猪PCV2特异性抗体水平稍高于二联疫苗免疫组,但二者无显著差异(P>0.05);二联疫苗免疫组与单独使用SVV疫苗免疫组仔猪SVV中和抗体效价也相当。首免后28 d相应实验组分别经滴鼻肌注各2 mL感染PCV2(10^(5)TCID50/mL)或SVV(10^(9.5)TCID50/mL),进行攻毒保护试验,结果显示二联疫苗免疫效果与PCV2单苗免疫效果相当,针对PCV2的攻毒保护率均为80%;针对SVV攻毒,二联疫苗免疫组及SVV单苗免疫组仔猪均无明显临床症状,保护率均为100%。以上结果表明PCV2-VLP和SVV全病毒灭活二联疫苗免疫效果与单独PCV2疫苗或SVV疫苗免疫效果相当,该灭活二联苗免疫仔猪后能够诱导机体产生体液免疫应答,具有良好的免疫保护作用。本研究首次将PCV2-VLP和SVV全病毒灭活疫苗进行联合免疫,试制了PCV2-VLP和SVV全病毒灭活二联疫苗,并评价了其对仔猪的免疫效力,该疫苗对临床预防PCV2及SVV感染具有潜在的应用价值。In order to evaluate the immunogenicity of bivalent porcine circovirus type 2 virus-like particles(PCV2-VLP)and inactivated seneca valley virus(SVV)vaccines,the immune efficacy of the bivalent vaccine made from PCV2-VLP and inactivated SVV vaccine,which prepared by mixing those two vaccines at equal volume and then combining with the ISA201 adjuvant,or two vaccines separately were evaluated in pigs.The results showed that the immunoprotective effect of the bivalent vaccine against PCV2 or SVV is similar with that of the PCV2 vaccine and SVV vaccine used alone.The immunoprotection against PCV2challenge was 80%in both PCV2 vaccine groups.And the bivalent vaccine showed complete protection against SVV challenge and the pigs in either of SVV vaccinated groups had no clinical signs after challenged with SVV.In addition,the piglet blood samples were collected at different time points after immunization with bivalent vaccine,PCV2 vaccine and SVV vaccine,separately.And the serum antibody levels of PCV2 and SVV were examined by indirect ELISA or virus neutralization test.The results showed that the PCV2-specific antibody in the PCV2 vaccine group is slightly higher than that in the bivalent vaccine group,but with no significant difference(P>0.05).As well as,the bivalent vaccine group induced similar SVV neutralizing antibody level in comparison of that in the SVV vaccine group.These results suggested that bivalent vaccine,PCV2-VLP vaccine and SVV vaccine could provide similar protective efficacy against PCV2 and SVV infection in swine.In general,the PCV2-VLP vaccine and inactivated SVV vaccine were used for the first time in combined immunization study.The bivalent vaccine was prepared and the immune efficacy was evaluated in piglets.The bivalent vaccine developed in this study has potential application value for the prevention against PCV2 and SVV infection.
