摘 要:为探究结缔组织生长因子CTGF对奶牛乳腺上皮细胞生长特性及其乳蛋白合成的影响,在细胞对数生长期,以无血清生长培养液为对照,添加CTGF(200 ng·mL^(-1))检测奶牛乳腺上皮细胞增殖和凋亡情况;在诱导细胞泌乳分化时,通过额外添加CTGF,检测β-酪蛋白mRNA和蛋白含量变化,评估其对乳蛋白的影响。结果表明,添加CTGF促进奶牛乳腺上皮细胞增殖,抑制其凋亡,CTGF可上调PRL/PRLR-STAT5和整联蛋白β1途径提高β-酪蛋白合成分泌。研究发现,CTGF作为一种生长因子,有利于乳腺上皮细胞生长存活,可正向调控重要泌乳分化信号途径,促进泌乳分化细胞乳蛋白合成,具备应用于体外泌乳调控的潜力。To investigate the effects of connective tissue growth factor(CTGF) on the growth characteristics and milk protein synthesis in bovine mammary epithelial cells, the proliferation and apoptosis of bovine mammary epithelial cells were detected after adding CTGF(200 ng·mL^(-1)) in serumfree growth medium during logarithmic growth period. The mRNA level of β casein and protein content were detected after adding additional CTGF during inducing lactating differentiation of cells, and the effects of CTGF on milk protein were evaluated. The results showed that adding CTGF could promote the proliferation and inhibit the apoptosis in mammary epithelial cells. And CTGF could up-regulated PRL/PRLR-STAT5 and integrin β1 pathways to enhance β casein synthesis and secretion. The results suggested that CTGF, as a growth factor, was beneficial to the growth and survival of mammary epithelial cells, and could positively regulate the important signaling pathways to lactation differentiation,and further promote the synthesis of milk protein in lactation differentiation cells, which had the potential to be applied for the regulation of in vita lactation.
