摘 要:目的观察苦参素通过调节电压门控性钾通道促进离体大鼠胰岛素分泌的效果。方法运用胶原酶P消化法和DispaseⅡ消化法分别得到大鼠的胰岛和胰岛β细胞。大鼠胰岛用于胰岛素分泌实验,采用放射免疫法,实验分为2.8 mmol/L葡萄糖组、2.8 mmol/L葡萄糖+苦参素(1μmol/L)组、2.8 mmol/L葡萄糖+苦参素(10μmol/L)组、2.8 mmol/L葡萄糖+苦参素(100μmol/L)组、11.1 mmol/L葡萄糖组、11.1 mmol/L葡萄糖+苦参素(1μmol/L)组、11.1 mmol/L葡萄糖+苦参素(10μmol/L)组、11.1 mmol/L葡萄糖+苦参素(100μmol/L)组、16.7 mmol/L葡萄糖组、16.7 mmol/L葡萄糖+苦参素(1μmol/L)组、16.7 mmol/L葡萄糖+苦参素(10μmol/L)组、16.7 mmol/L葡萄糖+苦参素(100μmol/L)组。胰岛β细胞用于膜片钳实验,测定电压门控性钾通道电流和动作电位,实验分为对照组和苦参素(10μmol/L)组。结果胰岛素分泌实验表明,低糖(2.8 mmol/L)时,苦参素(1μmol/L、10μmol/L、100μmol/L)未影响胰岛素的分泌(P>0.05)。高糖(11.1 mmol/L、16.7 mmol/L)时,苦参素(10μmol/L、100μmol/L)促进了胰岛素分泌(P<0.01)。膜片钳实验结果显示,苦参素(10μmol/L)抑制了电压门控性钾通道,延长了动作电位时程。结论离体大鼠的胰岛中,苦参素通过抑制电压门控性钾通道,延长了动作电位时程,促进了胰岛素分泌。Objective To explore the effect of oxymatrine on insulin secretion in isolated rats by regulating voltage-gated potassium channels.Methods Pancreatic islets and isletβcells of rats were obtained by collagenase P digestion and DispaseⅡdigestion.Pancreatic islets of rats were used for insulin secretion experiments by radioimmunoassay.The experiment were divided into 2.8 mmol/L glucose group,2.8 mmol/L glucose+oxymatrine(1μmol/L)group,2.8 mmol/L glucose+oxymatrine(10μmol/L)group,2.8 mmol/L glucose+oxymatrine(100μmol/L),11.1 mmol/L glucose group,11.1 mmol/L glucose+oxymatrine(1μmol/L)group,11.1 mmol/L glucose+oxymatrine(10μmol/L)group,11.1 mmol/L glucose+oxymatrine(100μmol/L)group,16.7 mmol/L glucose group,16.7 mmol/L glucose+oxymatrine(1μmol/L)group,16.7 mmol/L glucose+oxymatrine(10μmol/L)group,16.7 mmol/L glucose+oxymatrine(100μmol/L)group.The isletβcells were used for patch-clamp experiments to measure voltage-gated potassium channel currents and action potentials.The experiments were divided into control group and oxymatrine(10μmol/L)group.Results The results of insulin secretion experiment showed that oxymatrine(1μmol/L,10μmol/L,100μmol/L)did not affects on insulin secretion at low glucose(2.8 mmol/L)(P>0.05).At high glucose(11.1 mmol/L,16.7 mmol/L),oxymatrine(10μmol/L,100μmol/L)promoted insulin secretion(P<0.01).The results of patch-clamp experiments showed that oxymatrine(10μmol/L)inhibited the voltage-gated potassium channel and prolonged the action potential duration.Conclusion In isolated rat islets,oxymatrine prolonged the action potential duration and promoted insulin secretion by inhibiting voltage-gated potassium channels.
