摘 要:骨关节炎(OA)是一种临床慢性关节疾病,其病理改变主要为软骨退行性变,而社会老龄化的加重使得OA的发病率和致残率逐步攀升,成为困扰人类健康的社会问题之一。随着分子生物学的逐渐发展,OA生物标志物的研究日益引起国内外学者的关注。关节软骨的代谢贯穿OA病程始终,大量现有的生物标志物也发现于这一过程,这些均为下一步从软骨代谢角度,探索敏感性高、特异性强的生物标志物奠定了理论基础。目前发现,细胞的衰老破坏、基质稳态的失衡、炎症介质的表达和氧化应激等相关生物标志物均与OA发生发展关系密切,但软骨代谢异常导致OA发生发展的分子机制尚未完全清楚,因此应在研究关节软骨代谢的基础上进一步探索新的OA生物标志物。Osteoarthritis(OA)is a clinical chronic joint disease,and its pathological change is mainly cartilage degeneration.The aggravation of aging society makes the incidence and disability rate of OA gradually increase,which has become one of the social problems afflicting human health.With the gradual development of molecular biology,the study of OA biomarkers has attracted increasing attention of domestic and foreign scholars.The metabolism of articular cartilage runs through the course of OA,and a large number of existing biomarkers are also discovered in this process,which lays a theoretical foundation for the next step to explore biomarkers with high sensitivity and specificity from the perspective of cartilage metabolism.At present,it is discovered that biomarkers related to cell aging damage,imbalance of matrix homeostasis,expression of inflammatory mediators and oxidative stress are closely associated with OA development,but the molecular mechanism of the occurrence and development of OA caused by abnormal cartilage metabolism has not been fully understood.Therefore,novel OA biomarkers should be further explored based on the study of articular cartilage metabolism.
