摘  要:为探讨氯胺酮麻醉对发育期大鼠肝脏氧化应激和细胞焦亡的影响。7日龄SD大鼠连续注射氯胺酮,对照组注射生理盐水,检测大鼠肝脏病理组织学、肝功能指标、氧化应激水平和炎性细胞因子含量;Western blott和qPCR法检测焦亡相关蛋白和mRNA表达含量。结果表明,与对照组相比,氯胺酮组大鼠肝脏结构受损、肝功能指标降低、氧化应激和炎性细胞因子水平增加、细胞焦亡水平(NLRP3、ASC、caspase-1和GSDMD蛋白和mRNA)上调。可知,氧化应激和细胞焦亡参与氯胺酮诱导肝损伤的发病过程,为氯胺酮在兽医临床应用及肝损伤机制提供理论依据和参考。To investigate the effect of ketamine on oxidative stress and pyroptosis in liver of developing rats,seven-day-old SD rats were continuously injected with ketamine,and the control group was injected with normal saline,the histopathological changes,liver function indices,levels of oxidative stress and the contents of inflammatory cytokines in liver were detected;Western blot and qPCR were used to detect the expression of pyroptosis-related protein and mRNA.It was observed that the ketamine group had damaged liver structure,decreased liver function indices,increased levels of oxidative stress and inflammatory cytokines,and upregulated expression levels of pyroptosis(NLRP3,ASC,caspase 1 and GSDMD proteins and mRNA)as compared with the control group.The results indicated that oxidative stress and pyroptosis were involved in ketamine-induced liver injury,and provided a theoretical basis and reference for the clinical application of ketamine in veterinary medicine and the mechanism of liver injury.
