摘 要:目的本实验研究紫外线对高糖环境下视网膜Müller细胞糖基化终末产物受体(RAGE)基因表达的影响。方法配置糖基化终末产物(AGE)-牛血清蛋白(BSA),以浓度梯度的方式来干预细胞,将Müller细胞分为正常对照组、BSA组、25μg/ml AGE-BSA组、50μg/ml AGE-BSA组和100μg/ml AGE-BSA组,将细胞内的培养基换用添加了BSA以及预先配制好的25、50、100μg/ml的AGE-BSA无血清培养基,处理细胞44 h,然后将细胞放置于超净台上,紫外灯管(25 W)辐照。根据上述AGE-BSA给药浓度对RAGE的表达情况,选定100μg/ml的给药浓度。将细胞分为正常对照组、BSA组和AGEBSA组,分别处理8、16、24、48、72 h后收集细胞,然后将细胞放置于紫外灯管下(25 W)辐照。分析不同浓度AGE-BSA以及不同作用时间100μg/ml AGE-BSA对Müller细胞RAGE基因表达的影响。结果25μg/ml AGE-BSA组、50μg/ml AGE-BSA组和100μg/ml AGE-BSA组Müller细胞的RAGE基因的表达均高于正常对照组,差异具有统计学意义(P<0.05),并且随着AGE-BSA浓度的增加,RAGE基因的表达也可以逐渐的增高,在AGE-BSA浓度为100μg/ml时RAGE基因的表达可达到峰值(P<0.05)。经100μg/ml处理过的Müller细胞可以表达少量的RAGE基因,处理8、16、24、48、72 h后Müller细胞的RAGE基因表达均高于正常对照组,差异有统计学意义(P<0.05),在AGE-BSA浓度为100μg/ml,处理48 h时RAGE基因表达可达到峰值(P<0.05)。结论紫外线环境下,持续升高的血糖可使视网膜Müller细胞的AGE生成增多,并通过与其受体RAGE的相互作用,加速了DR的发生和发展。Objective To study the effects of ultraviolet rays on receptor for advanced glycation end products(RAGE) gene expression in Müller cells under high glucose environment. Methods Advanced glycation end products-Bovine serum albumin(AGE-BSA) was configured to intervene with the cells in a concentration gradient, respectively, and Müller cells were divided into normal control, BSA, 25 μg/ml AGE-BSA, 50 μg/ml AGE-BSA and 100 μg/ml AGE-BSA groups. The medium in the cells was replaced with pre-prepared 25 μg/ml,50 μg/ml and 100 μg/ml AGE-BSA serum-free medium supplemented with BSA for 44 h, and then the cells were then placed on an super-clean table and irradiated with a ultraviolet lamp(25 W). The administration concentration of 100 μg/ml was determined based on the above-mentioned AGE-BSA administration concentration on the expression of RAGE gene. The cells were divided into normal control group, BSA group and AGE-BSA group, and the cells were collected after processing for 8, 16, 24, 48 and 72 h, respectively, and then placed in a ultraviolet lamp(25 W) for irradiation. The effects of different concentrations of AGE-BSA as well as 100 μg/ml AGE-BSA with different action times on RAGE gene expression in Müller cells were analyzed.Results The expression of RAGE gene in Müller cells of 25 μg/ml AGE-BSA group, 50 μg/ml AGE-BSA group and 100 μg/ml AGE-BSA group were higher than that of normal control group, and the difference was statistically significant(P<0.05). With the increase of AGE-BSA concentration, the expression of RAGE gene can also be gradually increased, and the expression of RAGE gene can reach the peak at the AGE-BSA concentration of 100 μg/ml(P<0.05). Müller cells treated with 100 μg/ml could express a small amount of RAGE gene, and the expression of RAGE gene in Müller cells after treatment for 8, 16, 24, 48 and 72 h was higher than that in normal control, and the difference was statistically significant(P<0.05), and the RAGE gene expression could reach a peak at AGE-BSA concentration
