摘　　要：目的探讨α-细辛醚调控内质网应激对食管癌Eca-109细胞增殖及凋亡的影响。方法实验分为空白组和低、中、高剂量实验组。空白组给予不含药物的RPMI-1640培养基进行培养;低、中、高剂量实验组分别给予含25,50和100 mg·L^(-1)α-细辛醚溶液的RPMI-1640培养基进行培养。用噻唑蓝法检测细胞增殖率,用实时荧光定量聚合酶链反应法检测细胞中免疫球蛋白重链结合蛋白(BIP)、C/BEP环腺苷酸反应元件结合转录因子同源蛋白(CHOP)及葡萄糖调节蛋白78(GRP78)mRNA的表达水平。结果低、高剂量实验组和空白组的细胞增殖能力(OD值)分别为0.78±0.14,0.50±0.09和0.92±0.12,BIP mRNA相对表达量分别为7.68±2.08,3.27±0.94和13.07±2.35,CHOP mRNA相对表达量分别为1.22±0.33,2.39±0.63和0.91±0.30,GRP78 mRNA相对表达量分别为2.11±0.59,5.86±1.47和0.84±0.31。低、高剂量实验组的上述指标与空白组相比,差异均有统计学意义(均P<0.05)。结论α-细辛醚能通过调节内质网应激相关蛋白BIP、CHOP及GRP78表达,达到促进食管癌细胞凋亡、抑制增殖的作用。Objective To explore the effects ofα-asarone on the proliferation and apoptosis of esophageal cancer Eca-109 cells by regulating endoplasmic reticulum stress.Methods The experiment was divided into four groups:blank group(drug-free RPMI-1640 medium)and experimental-L,-M,-H groups(RPMI-1640 medium containing 25,50 and 100 mg·L~(-1)α-asarone).The cell proliferation rate was detected by methyl thiazolyl tetrazolium.The mRNA expression of immunoglobulin heavy chain binding protein(BIP)and C/EBP homologus protein(CHOP)and glucose-regulated protein 78(GRP78)was detected by reverse transcription-polymerase chain reaction.Results The cell proliferation abilities(OD value)in the experimental-L,experimental-H group and blank group were 0.78±0.14,0.50±0.09 and 0.92±0.12,the relative expression levels of BIP mRNA were 7.68±2.08,3.27±0.94 and 13.07±2.35,the relative expression levels of CHOP mRNA were 1.22±0.33,2.39±0.63 and 0.91±0.30,the relative expression levels of GRP78 mRNA were 2.11±0.59,5.86±1.47 and 0.84±0.31,respectively.The differences were statistically significant between experimental-L,-H groups and blank group(all P<0.05).Conclusionα-asarone can promote the apoptosis of esophageal cancer cells and inhibit proliferation by regulating the expression of BIP,CHOP and GRP78proteins.
