摘 要:目的研究家蝇抗菌肽17(AMP-17)和家蝇天蚕素18(C18)抗菌肽对肿瘤细胞的体外抗肿瘤活性。方法将人白血病细胞(K562、Hel)、人结肠癌细胞(DLD-1、Caco2)、人肝癌细胞(HepG2)、人喉癌细胞(HEP-2)、人肺癌细胞(A549)分为正常组(只加完全培养基)、阳性对照组(9.38,18.75,37.50,75.00和150.00μg·mL^(-1)盐酸阿霉素)、AMP-17实验组(60,80,100,120和140μg·mL^(-1) AMP-17)和C18实验组(10,20,40,60和80μg·mL^(-1) C18)。用细胞计数试剂盒-8(CCK-8)法测定肿瘤细胞增殖的抑制能力,计算半抑制浓度(IC_(50));用CCK-8法检测AMP-17和C18对人正常单核细胞(THP-1)的细胞毒性;用微量分光光度法测定AMP-17和C18对小鼠红细胞的半数溶血值(HC_(50))。结果AMP-17、C18均可抑制肿瘤细胞增殖,AMP-17对肿瘤细胞K562、Hel、DLD-1、Caco-2、HEP-2、HepG2和A549的IC_(50)值分别为(58.91±3.57),(80.81±1.16),(98.37±4.25),(88.16±3.46),(64.56±2.05),(70.03±1.78)和>150μg·mL^(-1);C18对上述肿瘤细胞的IC_(50)值分别为(26.80±0.20),(14.17±2.42),(13.59±2.48),(54.64±3.48),(36.30±0.23),(72.15±5.08)和(28.82±1.11)μg·mL^(-1)。AMP-17、C18和阳性药盐酸阿霉素对THP-1的IC_(50)值分别为(60.90±1.55),(32.60±1.21)和(8.51±0.81)μg·mL^(-1)。AMP-17和C18对红细胞的HC_(50)分别为>80和(44.41±5.45)μg·mL^(-1)。结论AMP-17、C18具有抗肿瘤活性,但C18溶血性较强。Objective To investigate the in vitro antitumor activity of Musca antimicrobial peptide 17(AMP-17)and cercropin 18(C18)antimicrobial peptides against tumor cells.Methods Human leukemia cells(K562,Hel),human colon cancer cells(DLD-1,Caco2),human liver cancer cells(HepG2),human laryngeal cancer cells(HEP-2),human lung cancer cells(A549)were divided into normal group(complete medium only),positive control group(Adriamycin hydrochloride:9.38,18.75,37.50,75.00 and 150.00μg mL^(-1)),AMP-17 experimental group(AMP-17:60,80,100,120 and 140μg.mL^(-1))and C18 experimental group(C18:10,20,40,60 and 80μg.mL^(-1)),cell counting kit-8(CCK-8)assay was used to determine the inhibitory capacity of tumor cell proliferation and calculate the half-inhibitory concentration(IC_(50)).Detection of cytotoxicity of AMP-17 and C18 on human normal monocytes(THP-1)by CCK-8 assay.Determination of hemihemolytic values(HC_(50))of AMP-17 and C18 on mouse erythrocytes by micro spectrophotometry.Results AMP-17 and C18 inhibited tumor cell proliferation.The IC_(50) values of AMP-17 and C18 on tumor cells K562,Hel,DLD-1,Caco-2,HEP-2,HepG2 and A549 were(58.91±3.57),(80.81±1.16),(98.37±4.25),(88.16±3.46),(64.56±2.05),(70.03±1.78),>150μg·mL^(-1)and(26.80±0.20),(14.17±2.42),(13.59±2.48),(54.64±3.48),(36.30±0.23),(72.15±5.08),(28.82±1.11)μg·mL^(-1).The IC_(50) values of MP-17,C18 and the positive drug Adriamycin hydrochloride against THP-1 were(60.90±1.55),(32.60±1.21)and(8.51±0.81)μg·mL^(-1).The HC_(50) of AMP-17 and C18 on erythrocytes was>80 and(44.41±5.45)μg·mL^(-1).Conclusion AMP-17 and C18 have anti-tumor activity,but C18 has strong hemolytic.
