摘  要:目的探索半夏泻心汤对3-脱氧葡萄糖醛酮(3DG)诱导的糖尿病前期大鼠的保护作用及机制。方法SD大鼠连续灌服50 mg·kg^(-1)3DG,14 d,建立糖尿病前期大鼠模型。将造模成功大鼠按体质量随机分为模型组和实验组,每组10只;另取10只正常大鼠作为正常组。正常组和模型组均按10 mL·kg^(-1)·d^(-1)剂量灌胃给予生理盐水;实验组灌胃给予相当于5 g·kg^(-1)·d^(-1)生药的半夏泻心汤,每组均连续灌服14 d。采用酶联免疫吸附法测定结肠和血清中蛋白质羰基化程度、活性氧(ROS)以及白细胞介素-1β(IL^(-1)β)含量。结果正常组、模型组和实验组的结肠蛋白质羰基化程度分别为(3.20±0.21),(4.31±0.25)和(3.72±0.23)nmol·mg^(-1);这3组结肠ROS表达量分别为(738.10±17.29),(1082.00±51.93)和(834.80±27.63)U·mg^(-1);这3组结肠IL^(-1)β表达量分别为(176.10±9.95),(237.70±16.08)和(200.10±11.59)ng·L^(-1)。这3组血清中蛋白质羰基化程度分别为(0.53±0.06),(0.73±0.06)和(0.61±0.06)nmol·mg^(-1);这3组血清中ROS表达量分别为(295.60±38.90),(446.70±39.86)和(331.40±10.13)U·mL^(-1);这3组血清中IL^(-1)β含量分别为(25.33±0.90),(29.83±2.44)和(25.93±1.02)ng·L^(-1)。模型组的上述指标与正常组相比,实验组的上述指标与模型组相比,差异均有统计学意义(均P<0.05)。结论半夏泻心汤对糖尿病前期的保护作用可能与其降低结肠和血清中蛋白质羰基化水平、ROS水平以及炎症因子含量有关。Objective To explore the protective effect and mechanism of Banxia Xiexin decoction on pre-diabetes rats induced by 3 DG.Methods SD rats were given 3 DG(50 mg·kg^(-1))for 14 days to establish prediabetic rat model.The successful modeling rats were randomly divided into model group and experimental group according to body weight,with 10 rats in each group.Another 10 normal rats were selected as the normal group.Normal group and model group were given normal saline by gavage with volume of 10 mL·kg^(-1)·d^(-1).The experimental group was given Banxia Xiexin Decoction equivalent to 5 g·kg^(-1)·d^(-1) by gavage.Each group was given gavage for 14 days.Then,the protein carbonylation,reactive oxygen specios(ROS)and interleukin-1β(IL^(-1)β)in the colon and serum in rats were determined by enzyme linked immunosorbent assay(ELISA).Results The protein carbonylation degrees of colon proteins in the normal group,the model group and the experimental group were(3.20±0.21),(4.31±0.25)and(3.72±0.23)nmol·mg^(-1),respectively.The expression levels of ROS in colon of the 3 groups were(738.10±17.29),(1082.00±51.93)and(834.80±27.63)U·mg^(-1),respectively.The colon IL^(-1)βcontents in the 3 groups were(176.10±9.95),(237.70±16.08)and(200.10±11.59)ng·L^(-1),respectively.The degree of protein carbonyl in serum of the 3 groups was(0.53±0.06),(0.73±0.06)and(0.61±0.06)nmol·mg^(-1),respectively.The expression levels of ROS in the 3 groups were(295.60±38.90),(446.70±39.86)and(331.40±10.13)U·mL^(-1),respectively.The serum levels of IL^(-1)βin the 3 groups were(25.33±0.90),(29.83±2.44)and(25.93±1.02)ng·L^(-1),respectively.The above-mentioned indexes,compared between model group and normal group,the difference of the factors were significant(all P<0.05);compared between experimental group and model group,the difference of the factors were significant(all P<0.05).Conclusion Banxia Xiexin decoction in the treatment of pre-diabetes may be related to its reduction of protein carbonylation level,ROS level and inflammato
