摘 要:目的观察德都红花-7味散对四氯化碳(CCl_(4))所致肝纤维化大鼠B淋巴细胞瘤-2(Bcl-2)和Bcl-2相关X蛋白(Bax)mRNA表达的影响。方法将40只SD雄性大鼠按体质量分层随机分为空白组、模型组、对照组和实验组,每组10只。用腹腔注射CCl_(4)的方法建立肝纤维化模型。空白组和模型组均灌胃给予等量蒸馏水;对照组灌胃给予21.0 mg·kg^(-1)水飞蓟宾;实验组灌胃给予0.3 g·kg^(-1)德都红花-7味散。4组大鼠每天给药1次,连续给药7周。用Masson染色法观察肝纤维化的情况,用实时荧光定量聚合酶链反应法检测大鼠肝组织中Bcl-2和Bax mRNA的表达水平。结果实验组、对照组、模型组和空白组的胶原百分比分别为(7.99±1.71)%,(8.82±2.63)%,(14.84±4.45)%和(0.80±0.29)%,Bcl-2 mRNA表达量分别为0.64±0.13,0.78±0.18,1.07±0.43和0.48±0.16,Bax mRNA表达量分别为0.95±0.10,0.98±0.26,1.04±0.29和1.27±0.36。实验组的胶原百分比和Bcl-2 mRNA表达量与模型组比较,差异均有统计学意义(均P<0.05)。结论德都红花-7味散能够明显减轻CCl_(4)所致慢性肝损伤,其机制与调节Bcl-2 mRNA表达相关。Objective To observe the effect of Deduhonghua-7 powder on mRNA expression of B-cell lymphoma-2(Bcl-2)and Bcl-2 assaciated X(Bax)in carbon tetrachloride(CCl_(4))-induced liver fibrosis rats.Methods Forty male SD rats were randomly divided into blank,model,control and experimental groups according to body weight,with 10 rats per group.The liver fibrosis model was established by intraperitoneal injection of CCl_(4).The blank and model groups were given distilled water intragastrically,the control group was given 21.0 mg·kg^(-1) silymarin and the experimental group was given 0.3 g·kg^(-1) Deduhonghua-7 powder intragastrically once a day for 7 weeks.The pathological change of liver tissue was observed by Masson staining.The reverse transcription-polymerase chain reaction was used to detect the influence of Bcl-2 and Bax mRMA expression in rat liver tissue.Results The percentages of collagen of experimental,control,model group and blank groups were(7.99±1.71)%,(8.82±2.63)%,(14.84±4.45)%and(0.80±0.29)%,the Bcl-2 mRMA expression was 0.64±0.13,0.78±0.18,1.07±0.43 and 0.48±0.16,the Bax mRNA expression was 0.95±0.10,0.98±0.26,1.04±0.29 and 1.27±0.36.Compared with the model group,percentages of collegen and Bcl-2 mRNA expression in experimental group had significant differences(all P<0.05).Conclusion Deduhonghua-7 powder can significantly reduce chronic liver injury caused by CCl_(4),and the mechanism is related to the regulation of the mRMA expression of Bcl-2.
