摘 要:目的研究党参远志散微乳提取液对阿尔茨海默症(AD)小鼠海马亚区中乙酰胆碱酯酶(AchE)、乙酰胆碱转移酶(ChAT)蛋白表达及乙酰胆碱(Ach)含量变化的影响,进而运用胆碱能学说探讨党参远志散微乳提取液对阿尔茨海默症模型小鼠学习记忆的影响。方法小鼠随机分为6组,对照组、模型组、空白微乳组、阳性药组、党参远志散水提液组、党参远志散微乳提取组,每组15只。除对照组腹腔给药生理盐水外,其余组均腹腔给药D-半乳糖及亚硝酸钠复制AD小鼠模型,对照组和模型组灌胃蒸馏水,其他组分别灌胃空白微乳、脑复康片、党参远志散水提液、党参远志散微乳提取液6周。采用免疫组织化学染色法检测海马各亚区AchE蛋白及ChAT蛋白表达;酶联免疫分析测定脑组织中Ach含量。结果与模型组比较,党参远志散微乳提取组小鼠海马CA1区、CA4区及齿状回AchE蛋白表达明显降低(P<0.01),CA3区AchE蛋白表达明显降低(P<0.05);CA1区、CA3区及齿状回ChAT蛋白表达明显升高(P<0.01),CA4区ChAT蛋白表达明显升高(P<0.05);小鼠脑组织中Ach含量明显升高(P<0.01)。结论党参远志散微乳提取液可显著降低海马各亚区AchE蛋白表达、提高海马各亚区ChAT蛋白表达并显著提高脑组织中Ach含量发挥胆碱能效应,增强神经突触兴奋性,进而改善小鼠学习记忆障碍。Objective To study the effect of Dangshen Yuanzhi Powder(党参远志散)microemulsion extract on the expressions of AchE and ChAT and the content of ACh in hippocampal subregion of Alzheimer's disease(AD)mice.Furthermore,to study the effect on learning and memory of AD mice by cholinergic theory.Methods Mice were randomly divided into control group,model group,blank microemulsion group,positive drug group,Dangshen Yuanzhi Powder water extract group and microemulsion extract group,with fifteen mice in each group.In addition to the control group,the other groups were intraperitoneally administrated with D-galactose and sodium nitrite to duplicate the AD mice model.Then,the control group and model group were given distilled water.The other groups were given blank microemulsion,Acetamide Pyrrolidone,Dangshen Yuanzhi Powder water extract and microemulsion extract for 6 weeks.The expressions of AchE and ChAT protein in each subregion of hippocampus were detected by immunohistochemical staining.Enzyme-linked immunoassay was used to determine the content of Ach in brain tissue.Results Compared with that of the model group,AchE protein expression was significantly reduced in hippocampal CA1,CA4 and DG regions(P<0.01),and significantly reduced in hippocampal CA4 regions(P<0.05).ChAT protein expression was significantly increased in hippocampal CA1,CA3 and DG regions(P<0.01)and significantly increased in hippocampal CA4 region(P<0.05).Ach content in brain tissue of mice was significantly increased in Dangshen Yuanzhi Powder microemulsion extract group(P<0.01).Conclusion Dangshen Yuanzhi Powder microemulsion extract can significantly reduce the expression of AchE protein and increase the expression of ChAT protein in hippocampal subregions,significantly increase ACh content in brain tissue,exert cholinergic effect,enhance the excitability of synapses,and then improve the learning and memory disorders of mice.
