摘 要:在实验室和工业领域中,快速而廉价地制备目的蛋白的方法对于促进蛋白质科学和工程的研究至关重要。使用基于包涵体的蛋白质表达系统是一种经典的方法,在上游过程中可以获得高产量,但是在下游过程中未正确折叠的蛋白质复性过程通常会导致产量显著降低。最具挑战性的是,蛋白质的复性条件因构象不同各有差异,需要个体化的针对性研究进行反复试验来提高蛋白质终产量。因此,为了将这种麻烦的折叠过程转变为常规的折叠过程,基于新技术和新材料的各种方法应运而生。文章首先简略地概述蛋白质折叠,总结包涵体变性的不同方式,重点对在复性过程中的三种思路进行了综述,并就蛋白质复性方面已报道的热点描述了方法发展背后的概念,以期提供一种通用的复性研究思路。The method of manufacturing the target protein fast and cheaply is crucial in laboratories and industrial fields to promote protein research.A traditional way is to use a protein expression system that relies on inclusion bodies.Although large yields can be achieved in the upstream process,the downstream renaturation of improperly folded proteins usually results in a considerable loss in yield.The most difficult aspect is that protein renaturation conditions differ depending on shape,necessitating specific targeted study and repeated trials to increase the final protein yield.Therefore,several approaches based on new technologies and new materials have arisen in order to convert this difficult folding process into a standard folding process.This article begins by providing a brief overview of protein folding,then summarizes the various methods of inclusion body denaturation,focuses on the three ideas in the renaturation process,and describes the concepts behind the method development for the reported hot spots in protein renaturation.It is envisaged that as a result of this,a general approach to refolding research would develop in the future.
