摘  要:为初步探究禽呼肠孤病毒(ARV)σC蛋白对天然免疫信号通路的影响,本研究将构建好的真核质粒pEF1α-HA-σC(实验组)及pEF1α-HA(对照组)转染DF1细胞进行表达,通过RT-qPCR检测不同时间点DF1细胞中天然免疫相关因子在转录水平的变化。检测后统计发现,TLR3、TLR7、MDA5、TLR15、TRIF、NF-κB、IRF7、MyD88、MAVS、IFN-α、IFN-β、IL-1β及IL-6天然免疫相关因子在转录水平上都发生了不同程度的上调或下调,提示这13种天然免疫相关因子在ARVσC蛋白引起的天然免疫中发挥着重要作用。研究结果及分析为进一步揭示ARVσC蛋白在宿主抗病毒的天然免疫反应中发挥的作用及ARV的致病机理提供了研究基础,为研制新型高效靶向抗ARV疫苗及疫苗佐剂提供了理论依据。In order to explore the influence of ARVσC protein on the innate immune signal pathway,the constructed eukaryotic plasmids pEF1α-HA-σC(experimental group)and pEF1α-HA(control group)were transfected into DF1 cells for expression.RT-qPCR was used to detect the changes in the transcription level of natural immune factors in DF1 cells at different time points.After the test,statistics found that TLR3,TLR7,MDA5,TLR15,TRIF,NF-κB,IRF7,MyD88,MAVS,IFN-α,IFN-β,IL-1βand IL-6 innate immune-related factors have been up-regulated or down-regulated,so it is suggested that these 13 innate immune-related factors play an important role in the innate immunity caused by the ARVσC protein.The results and analysis provide a research basis in the natural immune response of the host antiviral and the pathogenic mechanism of ARV,and provide a theoretical basis for the development of targeted anti-ARV vaccines and vaccine adjuvants.
