摘 要:为探究铁过量对雏鸡肝氧化应激和组织病理学变化的影响,将30只1日龄海兰褐雏鸡随机分为对照组(C组)和铁过量组(I组),铁过量组雏鸡于10日龄采取口服灌胃的方式,连续3 d给予含300mg/mL FeSO_(4)·7H_(2)O的生理盐水,每只1 mL。于铁过量后第7、14、21天,每组随机取5只雏鸡,观察肝大体病理变化并计算器官指数;采用石蜡组织切片、HE染色及普鲁士蓝染色观察肝病理变化;采用化学比色法检测肝丙二醛(MDA)、一氧化氮(NO)、谷胱甘肽过氧化物酶(GPx)、过氧化氢酶(CAT)和总抗氧化能力(T-AOC)。结果显示,雏鸡肝MDA和NO含量于铁过量后7~21 d均显著或极显著升高(P<0.05或P<0.01);CAT和GPx活力于铁过量后7~14 d均显著或极显著降低(P<0.05或P<0.01);T-AOC于铁过量后7~21d均显著下降(P<0.05);铁过量组雏鸡肝出现结缔组织增生、炎性细胞浸润、含铁血黄素沉积等病理学变化。结果表明,过量铁使雏鸡肝组织发生损伤性变化,发生氧化应激,氧化-抗氧化反应失衡。To investigate the effects of iron overload on oxidative stress and histopathological changes in chickens liver,thirty 1-day-old Hyline brown chickens were randomly divided into two groups:the control group and the iron overload group.Chickens in the iron overload group at 10-day-old age were given FeSO_(4)·7H_(2)O 300 mg/mL normal saline by oral gavage for three days,1 mL per chicken.Five chickens in each group were randomly selected and their livers were taken to observe gross pathological changes and calculate the organ index on the 7th,14th,and 21st days after iron overload.The pathological changes of the liver were observed by paraffin tissue section,HE staining and Perls staining,chemical colorimetric method was used to detect the liver malondialdehyde(MDA),nitric oxide(NO),glutathione peroxidase(GPx),Catalase(CAT)and total antioxidant capacity(T-AOC).The results showed that the contents of MDA and NO increased significantly or extremely significantly(P<0.05 or P<0.01)from 7 d to 21 d after iron overload.CAT and GPx activity decreased significantly or extremely significantly(P<0.05 or P<0.01)from 7 d to 14 d after iron overload.T-AOC decreased significantly(P<0.05)from 7 d to 21 d after iron overload.The liver of groupⅠhad pathological changes such as connective tissue hyperplasia,inflammatory cell infiltration and hemosiderin deposition.This indicated that iron overload caused damage to chicken livers with oxidative stress,resulted in oxidative-antioxidative reaction imbalance.
