摘 要:2019年5月至2020年7月期间,运用形态学和分子生物学方法对叶尔羌河流域(塔县河段)塔里木裂腹鱼肠道内寄生的2种线虫进行种类鉴定及系统发育分析。结果显示,采集到的线虫据其形态数据可初步鉴定为杆咽属和对盲囊属,用PCR扩增的受检样本JG1、JG2的COⅠmtDNA序列与GenBank中已知的Rhabdochona salgadoi的同源性最高,为89.9%;用PCR扩增的受检样本JD1、JD2的ITS1 rDNA序列与鲁道夫对盲囊线虫B的同源性最高,为99.27%;鉴定结果表明,塔里木裂腹鱼上寄生的2种线虫为杆咽线虫未定种和鲁道夫对盲囊线虫;利用最大似然法和贝叶斯法分别构建了2种线虫的分子系统发育关系,并进行分析,其中杆咽属线虫的不同种类均能形成一个单系,COⅠmtDNA序列能很好地作为分子标记对其进行分析,但由于目前公布的杆咽属线虫分子数据较少,尚需要很多数据有待补充;待测对盲囊属线虫(Contracaecum(s.l))与来自波兰、中国、意大利等地区的鲁道夫对盲囊线虫聚为一支,系统发育结果很好地佐证了该线虫为鲁道夫对盲囊线虫。这些实验结果也为杆咽属线虫和对盲囊属线虫的系统发育进化研究补充了相关资料。Species identification and phylogenetic analysis of two nematodes parasitized in the intestinal tract of Schizothorax biddulphi in the Yarkand River(Taxian Section)were conducted using morphological and molecular biological methods between May 2019 and July 2020.The results showed that the collected nematodes can be tentatively identified to Rhabdochona and Contracaecum,based on their morphological data.PCR amplification of the COI mtDNA sequences of the examined samples JG1 and JG2 showed the highest similarity to the known Rhabdochona salgadoi in Gen Bank with 89.9%.PCR amplification of the ITS1 rDNA sequences of the examined samples JD1 and JD2 showed the highest similarity with Contracaecum rudolphii B at 99.27%.The results showed that the two nematodes parasitized in S.biddulphi were Rhabdochona sp.and C.rudolphii.The molecular phylogenetic relationships of the two nematodes were constructed and analyzed using Maximum likelihood and Mr Bayes methods,respectively,in which the different species of Rhabdochona could form a monophyletic lineage and the COⅠmtDNA sequence could be well used as a molecular marker for its analysis,but much data is still needed to be added due to the small amount of molecular data published for Rhabdochona.The nematode of the genus Contracaecum(s.l)was clustered with C.rudolphii from Poland,China,Italy and other regions,and the phylogenetic results well corroborate that the nematode is the C.rudolphii.These experimental results also add to the information related to phylogenetic evolutionary studies of nematodes of Rhabdochona and Contracaecum.
