摘  要:[目的]采用正交试验法优选钩藤碱最佳提取工艺,确定钩藤药材最佳采收时期。[方法]以传统外观质量和钩藤碱含量为评价指标,在单因素试验基础上,以料液比、乙醇质量浓度和超声处理时间为影响因素,采用L_(16)(4^(3))正交设计优化提取工艺。通过检测不同采收时期钩藤碱含量的变化,确定最佳采收时期。[结果]钩藤碱最佳提取工艺为料液比1∶20,60%乙醇浸泡1 h,超声处理40 min,80℃回流2 h;当年10月—次年2月为钩藤药材的较佳采收时期。[结论]优选出的钩藤碱提取工艺设计合理、结果可靠;确定了钩藤药材的采收时期,为利用湖南钩藤进行饮片生产提供了理论依据。[Objective]The optimal extraction conditions for rhynchophylline from Uncaria tenuifolia were investigated by orthogonal tess method,the optimal harvesting season for crude drugs was confirmed.[Method]Taking traditional appearance quality and rhynchophylline content as evaluation indicators,on the basis of single factor experiment,taking the solid-liquid ratio,ethanol mass concentration and ultrasonic treatment time as influencing factors,L_(16)(4^(3))orthogonal design was adopted to optimize the extraction process.The optimum harvesting period was determined by detecting the changes of rhynchophylline content in different harvesting periods.[Result]The optimal extraction process of rhynchophylline was as follows:solid-liquid ratio of 1∶20,soaking in 60%ethanol for 1 h,ultrasonic treatment for 40 min,and reflux at 80℃ for 2 h.The better harvesting time for Uncaria tenuifolia medicinal materials was from the year October to February of the following year.[Conclusion]The selected extraction condition for rhynchophylline was reasonable and dependable;the harvesting season for Uncaria tenuifolia medicinal materials was confirmed and provided the theoretical basis for production of Chinese herbal medicine in Uncaria tenuifolia.
