摘 要:目的分离鉴定牛源金黄色葡萄球菌,并分析其生物学特性。方法采用无菌操作从猝死牛的心、肝、脾、肺、肾等病变脏器组织中取样,用接种环划线,接种至LB固体培养板,于37℃培养12 h;挑取单菌落,进行革兰染色、镜检,再经纯培养,获得分离菌株。对分离菌株进行Baird-Parker氏培养板培养特性检测、血浆凝固酶试验、生化鉴定、药敏检测、16S rRNA同源性分析及动物致病性试验,并对猝死牛的脏器进行病理组织检测。结果获得的分离菌株为革兰阳性球菌,且多呈葡萄串状排列;于Baird-Parker氏培养基上生长为黑色菌落;血浆凝固酶试验呈阳性;可分解海藻糖、麦芽糖和尿素等物质,不能分解塔格糖、七叶苷、甲基葡萄糖苷等物质。16S rRNA基因PCR产物序列与金黄色葡萄球菌16S rRNA片段一致,表明分离菌株为金黄色葡萄球菌,命名为TL。TL与大连分离株KF495200同源性最高(99.3%),且具有较强的致病性。猝死牛的多个器官出现了细胞坏死等病变情况。结论分离菌株为致病性金黄色葡萄球菌。本实验为当地牛猝死症的流行病学调查及综合防控奠定了基础。Objective To isolate,identify and analyze the biological characteristics of Staphylococcus aureus.Methods Samples were taken aseptically from the heart,liver,spleen,lung,kidney of cattle which died suddenly and streak-inoculated to LB solid culture medium with inoculation loop and cultured at 37℃ for 12 h,from which single colonies were subjected to microscopy with Gram staining then to pure culture.The obtained isolates were subjected to culture characteristics test on Baird-Parker medium,plasma coagulase test,biochemical test,drug sensitivity test,16S rRNA homology analysis and animal pathogenicity test.The organs of cattle which died suddenly were subjected to pathological examination.Results The isolated strain was Gram-positive cocci,which was mainly arranged in grape strings,grew into black colonies on Baird-Parker medium and positive in plasma coagulase test.Trehalose,maltose and urea were decomposed by the isolated strain,while Tagatose,esculin and methyl glucoside were not.The 16S rRNA sequence of the strain,named as TL,was consistent with that of Staphylococcus aureus,which showed a homology of 99.3% to S.aureus strain KF495200 isolated in Dalian and high pathogenicity,resulting in the pathological change such as necrosis in various organs of cattle which died suddenly.Conclusion The isolated strain was pathogenic S.aureus.The study laid a foundation of epidemiological investigation as well as comprehensive prevention and control of bovine sudden death.
