摘　　要：目的检测神经嵴干细胞标记(p75、MSX2)和自我更新标记(OCT4、NANOG)在人毛囊隆突区迁出细胞的表达,并进一步检测其增殖活性。方法取正常头皮组织,显微分离单个毛囊隆突,并体外培养,观察迁出细胞的生长情况;采用免疫荧光染色法检测人毛囊隆突区迁出细胞p75、MSX2、OCT4和NANOG的表达;采用BrdU标记免疫荧光染色检测细胞增殖活性。结果p75、MSX2、OCT4和NANOG在人毛囊隆突区迁出细胞中呈阳性表达。传代培养24 h、48 h、72 h后BrdU阳性率分别为(49.50±4.62)%、(33.43±10.43)%、(17.51±8.51)%,且在传代培养24 h时增殖能力较高。结论人毛囊隆突区存在神经嵴干细胞,且该细胞具有迁移、自我更新能力及较强的增殖活性。Objective To detect the expression of neural crest stem cell markers(p75,MSX2)and self-renewal markers(OCT4,NANOG)of the emigrated cells from the bulges of human hair follicles,and to further examine their proliferative activity.Methods The single hair follicles was isolated from the normal scalp tissues under microscope,and were cultured in vitro to observe the growth of the migrated cells from the hair follicles.The expression of p75,MSX2,OCT4 and NANOG in the migrated cells from human hair follicles were detected by immunofluorescence staining.The proliferation activity of the cells was detected by BrdU-labelled immunofluorescence staining.Results p75,MSX2,OCT4 and NANOG were positively expressed in neural crest stem cells migrating from the bulges of human hair follicles.The mean positive rate of BrdU staining after 24,48 and 72 hours of passaging culture was(49.50±4.62)%,(33.43±10.43)%and(17.51±8.51)% respectively,with a higher proliferation activity at 24 hours of passaging culture.Conclusion The neural crest stem cells were present in the bulges of human hair follicles,and the cells had the ability to migrate,self-renew and a strong proliferative activity.
