摘  要:目的:研究马钱苷(Loganin,Log)对结肠癌裸鼠的抑瘤作用,并探讨其作用机制。方法:通过腋窝下接种结肠癌细胞建立裸鼠肠癌动物模型评价马钱苷的药理活性,并对裸鼠体重、肿瘤体积、抑瘤率等指标进行检测。裸鼠血清与肿瘤组织细胞因子采用商品化试剂盒检测,裸鼠血清与肿瘤组织超氧化歧化酶(Superoxide Dismutase,SOD)、过氧化氢酶(Catalase,CAT)、谷胱甘肽过氧化物酶(Glutathione peroxidase,GSH-Px)、与丙二醛(Malondialdehyde,MDA)采用商品化试剂盒检测。肿瘤组织病理学采用苏木精-伊红(HE)染色,裸鼠肿瘤组织Nrf-2/NO-1信号与ROCK/NF-kB信号采用免疫组化与蛋白印迹方法进行检测。结果:结果显示,Log增加裸鼠体质量、减少肿瘤体积,降低SOD、CAT、GSH-Px水平,升高MDA水平,升高血清与肿瘤组织细胞因子水平,缓解肿瘤组织病理学改变。同时,Log还能显著抑制Nrf-2/NO-1与激活ROCK/NF-kB信号蛋白表达。结论:Log对裸鼠体内结肠癌具有抑制作用,其机制可能与抑制Nrf-2/NO-1以及增强ROCK/NF-kB信号通路有关。Objective:To investigate the mechanism of Loganin(Log)inhibiting colon cancer in nude mice.Methods:Animal model of colorectal cancer in nude mice was established by inoculating colon cancer cells under armpit to evaluate the pharmacological activity of loganin.The body weight,tumor volume and tumor inhibition rate of nude mice were detected.The serum of nude mice and tumor tissue cytokines were detected by commercial kits,while the serum of nude mice and tumor tissue superoxide dismutase(SOD),catalase(CAT),glutathione peroxidase(GSH-Px),and malondialdehyde(MDA)were detected by commercial kits.Nude mice tumor tissue Nrf-2/NO-1 signal and ROCK/NF-kB signal were detected by immunohistochemistry and western blot.Results:Our results showed that Log significantly increased the body weight of nude mice,decreased tumor volume,significantly decreased SOD,CAT,GSH-Px,increased MDA content,and significantly increased the cytokine content in serum and tumor tissue.In addition,Log can significantly inhibit Nrf-2/NO-1 and activate ROCK/NF-kB signaling protein expression.Conclusion:Log has obvious antitumor effect on colon cancer in nude mice,and its mechanism is related to inhibite Nrf-2/NO-1 and to enhance ROCK/NF-kB signals.
