摘 要:目的 研究长链非编码RNA(lncRNAs)在单发及多发性子宫肌瘤组织中的表达及临床意义。方法 收集我院妇科2016年1月至2020年12月单发、多发性子宫肌瘤手术切除标本(各100例),选取其肿瘤瘤心为肌瘤组,内膜下1cm子宫肌壁层为对照组。经查阅资料筛选出与子宫肌瘤相关但尚未报道的lncRNAs LINC00958、MIR4435、SNHG5,利用实时荧光定量聚合酶链式反应检测其在单发、多发性子宫肌瘤组和对照组标本中的表达,并比较其表达差异。结果 方差分析显示,LINC00958、MIR4435和SNHG5在单发性子宫肌瘤组、对照组、多发性子宫肌瘤组、对照组四组标本中表达量的差异均有统计学意义(F值分别是907.400、26.160、3.390,P<0.05)。进一步两两比较发现,LINC00958在单发、多发性肌瘤组中的表达量均低于其相应的对照组,差异有统计学意义(t值分别是79.690、32.080,P<0.05);而MIR4435、SNHG5在单发、多发子宫肌瘤组中的表达量与其相应对照组均无统计学差异(P>0.05)。结论 LINC00958在单发及多发子宫肌瘤标本中的表达量均显著低于其肌瘤旁组织,可能成为子宫肌瘤临床预测、诊断的重要分子标记物和未来治疗的靶点。Objective To study the expression and clinical significance of long noncoding RNAs(lncRNAs) in single and multiple uterine fibroids.Methods The surgical specimens of single and multiple uterine fibroids(with 100 cases in each diagnosis) were collected from January 2016 to December 2020 in department of gynecology of this hospital.The tumor heart was selected as the fibroids group, and the myometrial wall of the 1 cm under the endometrium was selected as the control group.After consulting the data, the lncRNAs related to uterine fibroids but not reported yet were screened, including LINC00958,MIR4435 and SNHG5.Their expressions in the single and multiple uterine fibroids group and the control group were detected by real-time fluorescence quantitative polymerase chain reaction, and the differences of their expressions were compared.Results Variance analysis showed that there were significant differences in the expressions of LINC00958,MIR4435 and SNHG5 in single uterine fibroids group and control group, multiple uterine fibroids group and control group(F=907.400,26.160 and 3.390,respectively, P<0.05).Further pairwise comparison showed that the expression of LINC00958 in single and multiple uterine fibroids groups was lower than those in the corresponding control groups, and the differences were statistically significant(t=79.690 and 32.080,respectively, P<0.05).However, there were no significant differences in the expressions of MIR4435 and SNHG5 between the single and multiple uterine fibroids groups and their corresponding control groups(P>0.05).Conclusion The expression of LINC00958 in the single and multiple uterine fibroids specimens was significantly lower than that in the adjacent tissues, which may become an important molecular marker for clinical prediction and diagnosis of uterine fibroids and a target for treatment in the future.
