摘 要:目的模拟临床表现建立银环蛇咬伤致肌无力SD大鼠模型。方法检测所用银环蛇毒的毒力,采用不同剂量银环蛇干毒给予SD大鼠皮下注射,观察SD大鼠运动、呼吸等情况,确定本实验所需模型的银环蛇干毒最佳剂量。模型SD大鼠与对照组SD大鼠行重复神经电刺激检查,并于建模后3 h取SD大鼠膈肌、腓肠肌行HE染色,了解肌肉细胞情况。结果通过预实验,本实验所用银环蛇干毒的KM小鼠半数致死量为0.12 mg/kg,SD大鼠的半数致死量为0.08 mg/kg,实验所需模型的银环蛇干毒的最佳剂量3倍SD大鼠半数致死量0.24 mg/kg,该模型中毒表现与临床表现符合,低频重复神经电刺激检查有肌电衰减,并出现膈肌及腓肠肌细胞肿胀、核排列紊乱的病理改变。结论用3倍SD大鼠半数致死量的银环蛇毒,可成功建立银环蛇咬伤致肌无力的SD大鼠模型,可用于救治银环蛇咬伤早期肌无力的基础研究。Objective To establish model of myasthenia induced by Bungarus multicinctus bite in Sprague-Dawley(SD)rats.Methods The virulence of bungarotoxin was detected.Different doses of bungarotoxin were subcuta-neously injected into SD rats.The optimal dose of bungarotoxin was determined with observing the movement and respira-tion of SD rats.The rats in model and the control groups were given with repeated nerve stimulation test;and the dia-phragm and gastrocnemius of SD rats were taken for HE staining 3 hours after modeling.Results The median lethal dose of Bungarotoxin was 0.12 mg/kg in KM mice and 0.08 mg/kg in SD rats.The optimal dose of bungarotoxin was 0.24 mg/kg,which was 3 times of the median lethal dose in SD rats.The poisoning manifestations of the model were showed symp-toms similar to the clinical.Low frequency repetitive nerve stimulation examination showed electromyographic attenuation in model rats.Diaphragm and gastrocnemius muscle cells were edematous while nuclei were disordered in model rats.Conclusion The SD rat model of myasthenia caused by Bungarus multicinctus bite was successfully established by using 3 times of median lethal dose of Bungarotoxin;which can be used for the basic research of curing early myasthenia induced by Bungarus multicinctus bite.
