摘  要:目的 观察经口摄入天麻的亚急性毒性,初步评估观察到有害作用最低水平和/或未观察到有害作用水平,为安全食用天麻提供依据。方法 研究选用80只SPF级初断乳SD大鼠,随机分为4组,每组20只,雌雄各半;设2.5 g、5.0 g和10.0 g/kg·BW 3个剂量组和空白对照组,剂量组自由进食拌有天麻的饲料,对照组给予维持饲料,持续28 d。观察动物中毒表现及死亡情况;试验开始前和结束时,对照组及高剂量组动物进行眼科检查;称量并记录动物体重、摄食量,计算每周食物利用率及总食物利用率;试验结束时,采集动物24h尿液标本及血液标本,进行尿常规、血常规、血生化指标检测;动物安乐死后,解剖观察脏器大体改变,称量脏器湿重,计算脏体比;取对照及高剂量组器官组织行组织病理学检查。结果 试验期间动物健康状况良好,未观察到明显中毒症状,无动物死亡。眼科检查未见异常。各剂量组雌雄大鼠初始体重、每周体重、每周食物利用率和总利用率与对照组比较差异均无统计学意义(P>0.05)。试验中SD大鼠体重均呈增加趋势,实验结束时各组体重、平均食物利用率与对照组比较差异无统计学意义(P>0.05)。各剂量组雌雄大鼠血细胞学及凝血因子检测结果与对照组比较,差异无统计学意义(P>0.05)。血生化分析结果显示,与对照组比较,中、高剂量组雄性SD大鼠总蛋白、白蛋白显著升高;中剂量组雄性SD大鼠谷丙转氨酶、血糖、K+显著升高;高剂量组雄性SD大鼠肌酐、Na+显著升高;低、中剂量组雌性SD大鼠血糖显著降低,甘油三酯升高;高剂量组雌性SD大鼠血清Na+、Cl-显著升高;差异均有统计学意义(P<0.05)。以上指标均在正常值范围内,差异无生物学意义。结论 天麻对SD大鼠无明显可见毒副作用;在本实验条件下,天麻对SD大鼠的亚急性毒性NOAEL为10.0 g/kg·BW。Objective To observe the sub-acute toxicity of gastrodia elata ingested orally,assess the lowest observed adverse effect level(LOAEL) and/or no observed adverse effect level(NOAEL),and to provide basis for safely edible gastrodia elata.Methods Eighty SPF Sprague-Dawley rats were randomly divided into three dose groups(2.5 g,5.0 g and 10.0 g/kg · BW) and the blank control group groups with equal sex ratio in each group.The dose groups were administrated the feed that mixed with gastrodia elata freely for 28 days,and the control group was given the maintenance feed.Poisoning performance and death of the animals were observed.Before and at the end of the experiment,the animals in control group and high-dose group received ophthalmology examination.Body weight and food intake of animals were recorded weekly,then the weekly food utilization rate and total food utilization rate were calculated.At the end of the experiment,24 h urine and blood samples were collected for routine urine detection,hemocytology and blood biochemical indexes tests.The animals were anesthetized and sacrificed,the gross changes of organs were observed,the wet weight of organs was measured,and the visceral-body ratio was calculated.Organs and tissues in the control group and the high-dose group were collected for histopathological examination.Results During the experiment,animals were in good health;not obvious symptoms and deaths were observed.No eye abnormalities were observed.There were no significant differences in baseline and weekly weight,weekly food utilization and total utilization between male and female rats in the dose group and the control group(P>0.05).Body weight of the SD rats were gradually increased during the test,and there were no significant differences in body weight and average utilization rate of the food compared with the control group at the end of the experiment(P>0.05).There were no significantly difference in hemocytology and blood coagulation factor analysis between the treatment and control group(P>0.05).Compared
