摘  要:目的探讨白藜芦醇对哮喘模型鼠气道炎症及JAK-STAT6信号通路的影响。方法将6~8周龄SPF级雌性BALB/c小鼠18只随机分为3组:A组:对照组;B组:哮喘组;C组:哮喘经白藜芦醇治疗组;每组6只。B、C组用卵清蛋白(OVA)构建哮喘模型。实验第21天起C组给予白藜芦醇(100 mg/kg)灌胃,A、B组给予生理盐水灌胃,每组连续1周。第28天处死小鼠,取小鼠右肺观察病理损伤情况,用阿新蓝/过碘酸雪夫染色(Periodic acid Schiff and Alcian blue stain,AB-PAS)法观察气道黏液分泌情况。ELISA法检测肺组织匀浆上清液中IgE、白细胞介素(IL)-4、IL-13表达,Western blot检测肺组织中信号传导及转录激活蛋白6(signal transducer and activator of transcription,STAT6)、p-STAT6的表达。结果与A组相比,B组小鼠气道炎症更重、黏液分泌增加,肺内IgE、IL-4、IL-13含量增加,p-STAT6的表达增强(均P<0.05),经白藜芦醇治疗后(C组)气道炎症反应及黏液分泌均减轻,肺组织匀浆上清液中OVA特异性IgE、IL-4、IL-13含量减少,p-STAT6的表达减少(均P<0.05)。结论白藜芦醇治疗可减轻哮喘小鼠气道炎症及黏液高分泌,降低IgE、IL-4、IL-13的表达,其机制可能是通过抑制JAK-STAT6通路活化。Objective To investigate the effects of resveratrol on airway inflammation and activation of JAK-STAT6 signaling pathway in asthmatic mice.Methods Eighteen SPF-grade female BALB/c mice(20-22 g)were randomly divided into 3 groups,Group A(blank control group),Group B(asthma group)and Group C(asthma and resveratrol group).Group B and C were sensitized and challenged with ovalbumin(OVA).On the 21^(th) day of the experiment,mice in Group C were intragastrically administered with resveratrol(100 mg/kg)for one week,while mice in Group A and B were intragastrically administered with normal saline instead.On the 28^(th) day,all the mice were sacrificed and the right lungs of mice were taken to evaluate the pathological injury.The secretion of airway mucus was evaluated by AB-PAS staining.The levels of IgE,IL-4 and IL-13 in the lung were measured by ELISA.The expression of signal transducer and activator of transcription 6(STAT6)and phosphorylated STAT6(p-STAT6)in the lung tissues were assessed by Western Blot.Results The airway inflammation of mice in asthmatic group was significantly enhanced compared with that of mice in normal control group.Mucus secretion,the levels of IgE,IL-4 and IL-13,and the expression of p-STAT6 were significantly increased in asthmatic mice as compared to those in control group(P<0.05),which were significantly reversed by treatment of resveratrol(P<0.05).Conclusion Treatment with resveratrol might protect OVA-induced asthmatic airway inflammation by inhibiting JAK-STAT6 pathway activation.
