摘 要:为了探究慢性冷暴露对小鼠产热及肝脏结构和脂质代谢相关蛋白表达的影响,本试验选取体重相近的8周龄C57BL/6小鼠36只,随机分为常温组和慢性冷暴露组,每组3个重复,每个重复6只小鼠。慢性冷暴露组进行每天4℃,连续随机3 h冷刺激,持续时间为21 d。冷暴露结束后全自动生化分析仪检测小鼠血清葡萄糖以及甘油三酯含量;苏木精-伊红(HE)染色观察小鼠肝脏组织形态;碘酸雪夫氏(PAS)染色检测小鼠肝糖原含量;蛋白免疫印迹法检测肝脏组织中脂质合成与分解相关蛋白表达水平。结果显示:与常温组相比,慢性冷暴露组小鼠血清中葡萄糖含量显著降低(P<0.05),甘油三酯含量显著降低(P<0.05);小鼠肝脏组织出现空泡样变,同时肝糖原含量显著减少(P <0.05);肝脏组织脂质合成蛋白固醇调节元件结合蛋白1(SREBP1)、脂肪酸合成酶(FASN)蛋白表达水平显著升高(P<0.05);脂质分解蛋白肉碱棕榈转移酶1(CPT1)、过氧化物酶体增殖物激活受体α(PPARα)的蛋白表达水平显著升高(P<0.05),肉碱棕榈转移酶2(CPT2)蛋白表达水平极显著升高(P<0.01)。综上所述,慢性冷暴露下血清葡萄糖、甘油三酯及肝糖原消耗增加,产热加快;肝脏组织出现稳态应激负荷范围内损伤;肝脏组织中脂质合成及分解相关蛋白表达增加。In order to explore the effects of chronic cold exposure on thermogenesis,liver structure and lipid metabolism related protein expression in mice,thirty-six 8-week-old C57BL/6 mice with similar body weight were randomly divided into normal temperature group and chronic cold exposure group. The rats in the chronic cold exposure group were subjected to cold stimulation at 4 ℃ every day for 3 h at random for 21 days. After cold exposure,the contents of glucose and triglyceride in serum were detected by automatic biochemical analyzer. The morphology of liver was detected by HE staining. The content of liver glycogen was detected by PAS staining. The expression levels of lipid synthesis and decomposition related proteins in liver tissue were detected by Western blot. The results showed that compared with the normal temperature group,the serum glucose and triglyceride contents in the chronic cold exposure group decreased significantly(P<0.05),the liver vacuolar degeneration and the liver glycogen content decreased significantly(P<0.05).The expression levels of lipid synthesis proteins sterol regulatory element binding protein 1(SREBP1)and fatty acid synthase(FASN)in the liver tissue significantly increased(P<0.05). The expression levels of lipid decomposing protein carnitine palmitoyl transferase 1(CPT1)and peroxisome proliferator activated receptor alpha(PPARα)increased significantly(P<0.05),and that of carnitine palmitoyl transferase 2(CPT2)increased significantly(P<0.01).The results show that under chronic cold exposure,the consumption of serum glucose,triglyceride and liver glycogen increase and the heat production accelerate;the liver tissue is injured under stable state stress load and the expression of lipid synthesis and decomposition-related proteins in liver increase.
