济川煎对阳虚型慢传输型便秘大鼠肠神经递质的干预作用

摘　　要：目的观察济川煎对阳虚型慢传输型便秘(STC)大鼠肠神经递质的影响,探究济川煎治疗阳虚型STC的作用机制。方法将40只SD大鼠随即分为正常组,模型组,莫沙必利组,济川煎组,采用复合病因法制备阳虚型结肠慢传输型便秘大鼠模型,分别给予莫沙必利、济川煎等药物治疗7 d,观察粪便干湿重比,测定大鼠肠推进功能,采用苏木素-伊红(HE)染色观察结肠组织病理学改变,Elisa法检测大鼠结肠中肠神经递质血管活性肠肽(VIP)、P物质(SP)、5-羟色胺(5-HT)水平。结果与模型组比较,济川煎组粪便干湿重显著降低(P<0.01),肠推进功能显著升高(P<0.01),济川煎组结肠中肠神经递质VIP显著降低(P<0.01),神经递质SP、5-HT显著升高(P<0.01)。结论济川煎可以显著改善阳虚型STC粪便干湿重比和肠推进功能,减轻结肠黏膜炎症损伤,其机制可能与其调节肠神经递质VIP、SP、5-HT的含量及表达相关。Objective To observe the effect of Jichuan(济川煎)on enteric neurotransmitters in rats with Yang-deficiency slow transit constipation(STC),and to explore the mechanism of Jichuan in the treatment of Yang-deficiency STC.Methods 40 SD rats were divided into normal group,model group,mosapride group,and Jichuan decoction group.The yang deficiency type colon slow transit constipation rat model was prepared by compound etiology method,and mosapride was given respectively.,Jichuanjian and other drugs were treated for 7 days,the dry-wet weight ratio of feces was observed,the intestinal propulsion function of rats was measured,hematoxylin-eosin(HE)staining was used to observe the pathological changes of colonic histopathology,and Elisa method was used to detect neurotransmitters in the colon in the midgut of rats The level of VIP,SP,5-HT.Results Compared with the model group,the dry and wet weight of feces in the Jichuan decoction group was significantly reduced(P<0.01),the intestinal propulsion function was significantly increased(P<0.01),and the enteric neurotransmitter VIP of the colon in the Jichuan decoction group was significantly decreased(P<0.01).The neurotransmitters SP and 5-HT increased significantly(P<0.01).Conclusion:Jichuanjian can significantly improve the fecal dry-wet weight ratio and intestinal propulsion function of Yang-deficiency STC,and reduce the inflammation of colonic mucosa.Its mechanism may be related to its regulation of the content and expression of enteric neurotransmitters VIP,SP and 5-HT.
