摘  要:为了探讨菠萝再生植株的遗传及表观遗传变异情况,本研究以‘台农21号’菠萝再生植株为试材,利用流式细胞技术(FCM)、简单重复序列(SSR)和甲基化敏感扩增多态性(MSAP)分析DNA序列、染色体倍性及DNA甲基化的变化。结果表明:再生植株存在叶刺、花序颜色等性状的变异。进一步分析结果表明,仅有1株出现了染色体非整倍加倍,其变异率为2.27%。检测到39条条带具有多态性,多态性比率为16.66%,其相似系数为0.84~1.00。再生植株DNA甲基化水平(总甲基化率53.53%~61.34%)低于母株(总甲基化率为67.90%),且同时存在甲基化(甲基化率7.50%~21.29%)和去甲基化(去甲基化率18.47%~39.05%)的模式,但以去甲基化为主。可见,组培环境能够诱导菠萝再生植株发生遗传及DNA甲基化的变异,但以DNA甲基化变异为主。本研究将为菠萝工厂化育苗及新种质的创制提供基础。In order to study the variation of genetic and epigenetic in regenerated plants from tissue culture,using flow cytometry(FCM),simple sequence repeat(SSR)and methylation sensitive amplified polymorphism(MSAP)analyzed the changed of DNA,chromosome ploidy and DNA methylation.The regenerated plants of’Tainong 21’were used as material in this study.The results showed that the regenerated plants had variation in leaf spines and inflorescence color.Further study results suggested that chromosome aneuploidy was found in only one regenerated plant,and the mutation rate was 2.27%.Polymorphism were detected in 39 bands,the polymorphism ratio was 16.66%,and the similarity coefficient ranged from 0.84 to 1.000.The DNA methylation level of regenerated plants(53.53%~61.34%)were lower than mother plant(67.90%).And both methylation(7.50%~21.29%)and demethylation(18.47%~39.05%)were presented in regenerated plants,moreover,the demethylation was the main pattern.It can be seen that tissue culture could induce variation of genetic and DNA methylation in pineapple regenerated plants,and the DNA methylation changed will be the main one.This study will provide a basis for industrialized seedling production and innovated germplasm of pineapple.
