摘 要:为了全面掌握广西境内海菜花野生居群分布的遗传多样性特点,本研究利用ISSR分子标记技术,结合NTSYS 2.10e软件、Popgen32软件对广西10个海菜花自然居群进行遗传多样性和亲缘关系研究。结果表明:筛选出的19条ISSR引物共扩增产生71条条带,其中56条为多态性条带,多态性比率为78.87%。10个海菜花居群的多态位点百分率为58.33%~71.15%,其中靖西市古文乡居群(GW)的多态性百分率最高,为71.15%;永福县百寿镇居群(YF)的多态性百分率最低,为58.33%。遗传多样性分析显示,10个海菜花居群的等位基因数(Na)平均为1.7887;有效等位基因数(Ne)平均为1.4056;Nei’s基因多样性指数(He)平均为0.2515;Shannon信息指数(I)平均为0.3873。多态性百分率、Na、Ne、He、I值均表明广西境内海菜花居群间的遗传多样性处于较低水平。基于Jaccard’s遗传相似系数的UPGMA聚类分析表明,靖西市庞陵村居群(PL)和德保县三合村居群(SH)的遗传关系最近,两个居群的遗传相似系数高达0.9437;鹿寨县中渡镇居群(LZ)和永福县百寿镇居群(YF)划分为一组,与其他8个地理居群的遗传关系稍远。聚类结果表明不同地理居群海菜花的亲缘关系与地理分布密切相关,研究结果为保护这一珍稀濒危物种提供了分子生物学依据和理论基础。In order to fully understand the genetic diversity of the wild distribution of Ottelia acuminata populations in Guangxi.We used ISSR molecular marker technology,along with NTSYS 2.10e software and Popgen 32 software to explore the genetic diversity and genetic relationship of 10 natural population distributions of Ottelia acuminata in Guangxi.A total of 71 bands were amplified from 19 ISSR primers,of which 56 were polymorphic bands with a polymorphism ratio of 78.87%.And 10 groups had found a higher percentage of polymorphic loci,ranging from 58.33%in Baishou Town,Yongfu County,to the highest 71.15%in Guwen Township(GW),Jingxi City.Genetic diversity analysis was reflected in Polymorphism percentage,Na,Ne,He,and I.The average of alleles(Na)in the 10 populations was 1.7887,the effective alleles(Ne)being 1.4056,and the average Nei’s gene diversity index(He)0.2515,together with Shannon information Index(I)averages 0.3873.All the above valuables indicated that the genetic diversity among the 10 groups was low.The UPGMA cluster analysis based on Jaccard’s genetic similarity coefficient revealed that the genetic relationship between Pangling village cluster(PL)and Sanhe village cluster(SH)in Debao county in Jingxi city is recent.The coefficient genetic similarity of the two populations is as high as 0.9437.The cluster(LZ)of Zhongdu town in Luzhai county and the cluster(YF)of Baishou town in Yongfu county are classified into one group,which is slightly distant from the other eight geographical population groups.The results of clustering suggested that the genetic relationship of Ottelia acuminata was closely related to geographical distribution.The study result could be served as a molecular biological basis and theoretical basis to protect this rare and endangered species.
