摘 要:以大花蕙兰‘爱神’(Cymbidium Lucky Gloria‘Aguri’)与莲瓣兰‘小雪素’(Cymbidium tortisepalum‘Xiao Xuesu’)杂交果荚为材料对其无菌萌发、增殖分化、生根等培养各阶段进行了初步研究,探索其组培快繁的最佳培养基配方。结果表明:无菌萌发以1/2 MS基本培养基为佳,适宜的NAA浓度为0.2 mg/L,可得到原球茎。在增殖分化培养中,添加NAA、6-BA组合比添加单一NAA效果佳,NAA对增殖影响较小。增殖培养适宜的培养基配方为1/2 MS+0.4 mg/L NAA+0.1~0.5 mg/L 6-BA。生根培养以1/2 MS为基本培养基,单纯添加1.0 mg/L NAA,生根率高、根数多、根长长,生根效果好。建立大花蕙兰‘爱神’与莲瓣兰‘小雪素’杂交种子组培快繁技术,可满足规模化和产业化栽培的需要,也为杂交兰的种质创新提供帮助。The hybrid pods of Cymbidium Lucky Gloria‘Aguri’and Cymbidium tortisepalum‘Xiao Xuesu’were used as the material for the preliminary study of its aseptic germination,proliferation differentiation,rooting and other culture stages,to explore the optimal medium formula for its tissue culture and rapid propagation.The results showed that 1/2 MS basic medium was the best medium for sterile germination,and the appropriate NAA concentration was 0.2 mg/L,and the protocorms were obtained.The effect of NAA and 6-BA on the proliferation and differentiation of hybrid orchids was better than that of single NAA.The suitable medium for proliferation culture was 1/2 MS+0.4 mg/L NAA+0.1~0.5 mg/L 6-BA.The rooting culture was based on 1/2 MS medium with 1.0 mg/L NAA.The rooting rate was high,the number of roots was large and the root length was longer.It is easy to produce satisfactory rooting effects.The establishment of tissue culture and rapid propagation technology of hybrid orchid Cymbidium Lucky Gloria‘Aguri’and Cymbidium tortisepalum‘Xiao Xuesu’can meet the needs of large-scale and industrial cultivation,and also provide help for the germplasm innovation of hybrid orchid.
