小麦肉桂醇脱氢酶基因(TaCAD1)全长cDNA序列及生物信息学分析

摘  要:小麦(Triticum aestivum)是世界三大主粮之一,倒伏严重制约了小麦产量的提高。小麦肉桂醇脱氢酶基因(TaCAD1)是小麦木质素合成的关键基因,小麦体内木质素含量越高,茎秆强度越高,抗倒伏能力随之增强。为了探明小麦肉桂醇脱氢酶基因的生物学功能,对TaCAD1基因编码的氨基酸序列进行生物信息学分析。结果表明:TaCAD1基因cDNA序列全长1311bp,包含1个长度为1083bp的开放阅读框(ORF),编码360个氨基酸。TaCAD1蛋白等电点(pI)为5.74,分子量为38625.45 Da,其不稳定系数为23.91,属于稳定蛋白质。推导的TaCAD1蛋白为疏水性蛋白,不存在跨膜区,可能位于细胞质内,该蛋白的二级结构元件主要为无规卷曲。TaCAD1蛋白在进化过程中较为保守,与节节麦、大麦、高羊茅、黑麦草、短花药野生稻CAD蛋白的一致性达94.81%。研究结果为进一步探究TaCAD1在小麦抗倒伏过程中的作用及小麦抗倒伏的分子育种提供参考。Wheat(Triticum aestivum)is one of the world's three major staple foods,and wheat lodging has severely restricted the increase in wheat production.The wheat cinnamyl alcohol dehydrogenase gene(TaCAD1)is a key gene for wheat lignin synthesis.The higher the lignin content in wheat,the higher the stalk strength and the en⁃hanced lodging resistance.In order to ascertain the biological function of wheat cinnamyl alcohol dehydrogenase gene,this paper conducts bioinformatics analysis on the amino acid sequence encoded by TaCAD1 gene.Bioinformat⁃ics analysis showed that the full-length cDNA sequence of TaCAD1 was 1311 bp,including an open reading frame(ORF)of 1083 bp in length,encoding 360 amino acids.The isoelectric point(pI)of TaCAD1 protein is 5.74,the molecular weight is 38,625.45 Da,and its instability coefficient is 23.91,which is a stable protein.The deduced TaCAD1 protein is a hydrophobic protein,there is no transmembrane region,and may be located in the cytoplasm.The secondary structure of the protein is mainly random coils.The TaCAD1 protein is relatively conservative in the evolutionary process,and it is 94.81%consistent with the CAD protein of Jiejie wheat,barley,tall fescue,ryegrass,and short anther wild rice.This study provides new ideas for further exploring the role of TaCAD1 in the process of wheat lodging resistance and molecular breeding of wheat lodging resistance.
