摘  要:以茵芋花粉为材料,采用TTC染色法、I_(2)-KI染色法、红墨水染色法、离体萌发试验法进行花粉生活力测定,并测定直接贮藏与简单预处理后的贮藏花粉活力的变化。结果表明:TTC染色法、I_(2)-KI染色法不适合于茵芋花粉生活力测定,红墨水染色法、离体萌发试验法差异不具有统计学意义,均可适于茵芋花粉生活力测定。茵芋花粉在贮藏前须经一定预处理,降低花粉含水量;7 d内贮藏可经预处理后室温进行,30 d内贮藏可于普通冰箱内进行,更长时间的贮藏可考虑低温甚至超低温的方式。Using Skimmia reevesiana pollen as material,using TTC dyeing method,I2-KI dyeing method and red ink dyeing method,and in vitro germination test method for determination of pollen viability,and the changes of pollen viability after direct storage and simple pretreatment were determined.The results showed that TTC dyeing method,I2-KI dyeing method was not suitable for the determination of Skimmia reevesiana pollen viability assay,red ink dyeing method,the difference from the germination test was not significant,can be suitable for the determination of Skimmia reevesiana.pollen viability.Skimmia reevesiana pollen should be pretreated before storage to reduce its moisture content;it can be stored at room temperature after pretreatment within 7 days,in ordinary refrigerator within 30 days,and low temperature or even ultra low temperature can be considered for longer storage.
