摘 要:胚胎发育晚期丰富蛋白-2(late embryogenesis abundant proteins-2,LEA-2)基因家族在植物生长发育和逆境胁迫中起重要作用。为了探究玉米(Zea mays)LEA-2家族蛋白对干旱胁迫的响应,本研究对玉米、高粱(Sorghum bicolor)和拟南芥(Arabidopsis thaliana)中的LEA-2进行蛋白序列比对,对ZmLEA-2家族基因进行了基因结构、保守基序的分析,及其在染色体上的位置及玉米种间的共线性分析,同时进行了玉米、高粱和拟南芥中LEA-2基因的共线性分析;依据转录组数据对ZmLEA-2家族基因进行了表达模式分析,并对ZmLEA53的功能进行了初步验证。结果表明:玉米中共鉴定出81个ZmLEA-2基因,在10条染色体上不均匀地分布;系统进化树分析表明,81个ZmLEA-2基因可分为7组,同一组基因间具有相似的基因结构及保守基序个数和排序。表达模式分析显示,81个ZmLEA-2基因呈现不同的表达模式。干旱胁迫下,ZmLEA53、ZmLEA7和ZmLEA64等基因表达量上升,复水后表达量下降,推测这些基因参与了玉米耐旱性调控。ZmLEA53的过表达可以提高酵母(Saccharomyces)菌株在干旱(PEG)、盐和高温胁迫下的生存率。研究结果为探索ZmLEA-2基因家族功能及解析其在干旱胁迫响应中作用途径提供参考。Late embriogenesis abundant protein-2(LEA-2)gene family play important role in plant growth,development and abiotic stresses.In order to investigate the response of ZmLEA-2 family proteins to drought stress,the LEA-2 protein sequence in maize(Zea mays),Sorghum bicolor,and Arabidopsis thaliana were aligned,also analyzed the gene structure,conserved motifs and position on the chromosome.The collinearity of ZmLEA-2 gene family in maize species were analyzed.Collinearity analysis between the maize,sorghum and Arabidopsis thaliana were also performed.The expression pattern of ZmLEA-2 family genes were analyzed based on the transcriptome data,and the function of ZmLEA53 was preliminary verificated.The results showed that 81 ZmLEA-2 genes were unevenly distributed on 10 chromosomes;the phylogenetic tree analysis showed that 81 ZmLEA-2 genes were divided into 7 groups,and genes in the same group had similar gene structure and conserved motifs.81 ZmLEA-2 genes exhibited different expression patterns.Under drought stress,the expression of ZmLEA53,ZmLEA7 and ZmLEA64 was up-regulated and decreased after rewatering.It was speculated that these genes were involved in the regulation of drought tolerance.qPCR results showed that the expression of ZmLEA53 was positively regulated by drought,salt and high temperature.Overexpression of ZmLEA53 could improve the survival rate of yeast(Saccharomyces)cells under drought(PEG),salt and high temperature stress,respectively.The results provide a basis for further functional identification of ZmLEA-2 gene family under drought stress.
