摘　　要：目的研究不同消毒方法对流感病毒RNA的消除效率,筛选可用于环境中核酸污染清除的有效方法。方法将纯化的流感病毒RNA滴染在玻片上作为研究对象,经消毒作用后,采用荧光定量PCR评价不同消毒方式对流感病毒RNA的消除效果。结果中和剂鉴定试验显示中和剂、中和产物对PCR反应体系无显著影响。消毒剂对流感病毒RNA的清除试验显示,500 mg/L含氯消毒剂、500 mg/L二溴海因分别作用10 min后,RT-PCR检测结果均为阴性。5种过氧化物类消毒剂及二氧化氯作用30 min,Ct值小幅度升高。碘伏、乙醇、季铵盐消毒剂及紫外线照射消毒,对流感病毒RNA的清除效率很弱。结论含氯消毒剂及二溴海因对流感病毒RNA具有较好的消除效果。Objective To study the efficacy of various disinfection methods on the elimination of influenza virus RNA,and screen the effective methods which can be used to eliminate the pollution of nucleic acid in the environment.Methods Purified influenza virus RNA was dropped on glass slide.Fluorescence quantitative PCR was used to evaluate the effect of different disinfection methods on the elimination of viral RNA after disinfection.Results The neutralizer identification test showed that no significant effect on PCR reaction was observed with the neutralizer and the neutralized product.The removal test of influenza virus RNA after disinfection showed that the PCR test was negative for the chlorine disinfectant of 500 mg/L and dibromohydantoin acted of 500 mg/L for 10 minutes.After disinfection of five peroxide disinfectants and one chlorine dioxide for 30 minutes respectively,the Ct value revealed partial removal.Iodophor,ethanol and quaternary ammonium salt showed very weak in removing influenza virus RNA,as well as ultraviolet radiation.Conclusion The chlorine disinfectant and dibromohydantoin have good effect on the elimination of influenza virus RNA,while other methods of disinfection are difficult to remove it completely.
