摘 要:本研究以肝脏出血和脾脏坏死为特征的病死鸭肝脏和脾脏组织接种SPF鸡胚进行病毒分离,并利用PCR方法进行病原鉴定,对分离病毒进行了回归雏鸭试验和S1基因测序分析。结果显示:该分离病毒为新型鸭呼肠孤病毒,并命名为NDRV SY株;将该分离病毒接种SPF鸡胚、BHK-21和Vero细胞进行传代培养,结果表明其能稳定适应鸡胚增殖和致鸡胚死亡,在BHK-21和Vero细胞两种传代细胞上也能良好增殖,并产生明显细胞病变。本研究为NDRV的疫苗和致病机制研究奠定了基础。In this study,affected tissues of the diseased ducks characterized by liver hemorrhage and spleen necrosis were inoculated into SPF chicken embryos for virus isolation and the isolated virus was analyzed by PCR,animal regression test and sequencing of S1 gene.The results showed that the isolated virus was a Novel duck reovirus designated as NDRV SY strain.The NDRV SY strain was passaged continuously in SPF chicken embryos,BHK-21 and Vero cells.The virus gradually adapted to grow in chicken embryos and caused death.It also propagated well on cell cultures and produced obvious cytopathy.Availability of the NDRV SY strain provided materials for the research of pathogenesis and vaccine development.
