摘 要:通过第二代高通量测序手段对枯叶蝶的基因组进行Survey分析,研究枯叶蛱蝶的起源、进化及适应性,明确适合该物种全基因组深度测序和组装策略.通过低覆盖度测序预测枯叶蝶基因组大小,并利用生物信息学方法估计该种的杂合率、重复序列和GC含量等基因组信息.结果显示:(1)枯叶蛱蝶中华亚种(Kallima inachus chinensis Swinhoe)的基因组大小估计为545.05 Mb, GC含量为35.24%,测序深度为52×;(2)除略小于Maniola jurtina (614.74 Mb)和翠袖锯眼蝶(Elymnia shypermnestra)(566.93 Mb)外,明显大于其余蛱蝶的基因组;(3)K-mer分析(K=17)结果显示枯叶蝶基因组具有明显的杂合峰,杂合率为1.22%,重复序列比例为48.03%.In order to further study the origin, evolution and adaptability of the orange dead leaf butterfly Kallima inachus chinensis Swinhoe, and to determine the sequencing strategy suitable for the whole genome, the genome survey of orange dead leaf butterfly was sequenced.Low coverage genomic survey sequencing was performed at first, and then large-scale whole genome deep sequencing was done.Using the next generation high-throughput sequencing technique as the research method, the genome size of orange dead leaf butterfly was determined, and the genomic information such as heterozygosity, repeat sequence and GC content were estimated by bioinformatics method.The results show that:(1) the genome size of the orange dead leaf butterfly is estimated to be 545.05 Mbp, the GC content is 35.24%, and the sequencing depth is 52×;(2) except for Maniola jurtina(614.74 Mb) and Elymnia shypermnestra(566.93 Mb), the genome is significantly larger than that of other nymphalids;(3) from the K-mer distribution curve, it is found that there is an obvious heterozygous peak in the genome of orange dead leaf butterfly, the heterozygosity rate is 1.22%, and the proportion of repetitive sequences is 48.03%.
