摘 要:目的 探讨黄芪多糖(Astragalus polysaccharide,APS)对二硝基氟苯(Dinitrofluorobenzene,DNFB)诱导的小鼠迟发型超敏反应(Delayed-type hypersensitivity,DTH)的影响。方法 雄性6~8周龄ICR小鼠50只,SPF级,体重(20±2)g,将小鼠随机分成空白对照组(NC组)、模型对照组(MC组)、地塞米松组(DEX组)、黄芪多糖低剂量组(APS-L组)以及黄芪多糖高剂量组(APS-H组),每组10只小鼠;利用背部涂抹DNFB建立DTH模型;DEX组、APS-L组、APS-H组,分别灌胃地塞米松2.5 mg/(kg·d)、APS 200 mg/(kg·d)、APS 400 mg/(kg·d)共7 d;NC组、MC组无菌三蒸水等体积灌胃7 d。MC组、DEX组、APS-L组、APS-H组末次给药0.5 h后,耳部涂抹DNFB。24 h后,检测小鼠耳肿胀和脾指数的变化;耳片组织做苏木素-伊红(Hematoxylin Eosin,HE)染色,光镜下观察小鼠耳片形态结构的变化;实时荧光定量PCR实验检测脾脏组织中干扰素-γ(Interferon-γ,IFN-γ)mRNA的表达水平;选择性肠道培养基活菌培养计数法检测小鼠肠道菌群的变化。结果 与模型组相比,APS-L组和APS-H组小鼠耳肿胀显著减轻(P<0.0001),脾指数明显下降(P<0.05);组织病理学结果显示黄芪多糖给药可减轻模型小鼠局部水肿及血管扩张,减少淋巴细胞浸润;实时荧光定量PCR结果显示,与MC组相比,APS-L组和APS-H组小鼠脾脏中IFN-γ的mRNA水平明显下降(P<0.0001);肠道培养基活菌培养计数结果显示,与NC组相比,MC组小鼠肠道大肠杆菌和肠球菌的数量明显增加(P<0.001),乳酸杆菌和双歧杆菌的数量明显减少(P<0.0001),提示MC组存在较明显的肠道菌群失调,与MC组相比较,APS-H组小鼠肠道大肠杆菌和肠球菌的数量明显减少(P<0.0001),乳酸杆菌和双歧杆菌的数量明显增加(P<0.0001)。结论 黄芪多糖具有抑制迟发型超敏反应的作用,该作用可能与抑制IFN-γ和调节肠道菌群有关。Objective To investigate the effect of astragalus polysaccharides(Astragalus polysaccharide, APS) on delayed-type hypersensitivity(Delayed-type hypersensitivity, DTH) induced by dinitrofluorobenzene(Dinitrofluorobenzene, DNFB) in mice.Methods 50 male ICR mice aged 6-8 weeks, SPF grade, weighing(20±2) g, were randomly divided into blank control group(NC group), model control group(MC group), dexamethasone group(DEX group), low-dose astragalus polysaccharide group(APS-L group) and high-dose astragalus polysaccharide group(APS-H group), with 10 mice in each group. The DTH model was established by smearing DNFB on the back. DEX group, APS-L group and APS-H group were gavaged with dexamethasone 2.5 mg/(kg·d), APS 200 mg/(kg·d) and APS 400 mg/(kg·d) respectively for 7 days. The ears of MC group, DEX group, APS-L group and APS-H group were smeared with DNFB 0.5 hours after the last administration. After 24 hours, the changes of ear swelling and spleen index were detected, the ear slices were stained with HE(Hematoxylin Eosin, HE), and the morphological changes of ear slices were observed under light microscope. The expression of Interferon-γ(Interferon-γ, IFN-γ) mRNA in spleen was detected by real-time quantitative PCR assay, and the changes of intestinal flora were detected by selective intestinal culture. Results Compared with the MC group, the ear swelling and spleen index of mice in APS-L group and APS-H group decreased significantly(P<0.0001), and the histopathological results showed that astragalus polysaccharides could reduce local edema, vasodilation, and lymphocyte infiltration in model mice. The results of real-time quantitative PCR showed that the mRNA level of IFN-γ in spleen of APS-L group and APS-H group was significantly lower than that of MC group(P<0.0001). The results of living bacteria culture in intestinal culture medium showed that compared with NC group, the number of Escherichia coli and Enterococcus in MC group increased significantly(P<0.001), while the number of Lactobacillus and Bifido
