摘  要:目的建立HPLC法同时测定黄瑞香注射液原料中瑞香素、伞形花内酯、芹菜素和芫花素的含量。方法采用Luna C;(2)100 A(250 mm×4.6 mm,5μm)色谱柱,以体积分数为0.05%的磷酸水溶液为流动相A,乙腈为流动相B,进行梯度洗脱。检测波长为326 nm。进样量20μL,柱温为35℃,流速1.0 mL·min^(-1)。结果瑞香素、伞形花内酯、芹菜素和芫花素色谱分离良好,分别在3.0~300.0、3.0~300.0、2.0~200.0和2.0~200.0 mg·L^(-1)内与峰面积呈良好的线性关系,r均大于0.9990;平均加样回收率分别为99.2%、98.7%、98.1%和99.4%;RSD分别为1.2%、1.7%、1.6%和1.3%(n=6)。结论建立的方法适用于研究黄瑞香注射液原料中瑞香素、伞形花内酯、芹菜素和芫花素的含量测定提供依据。Objective To establish a method for the determination of the contents of daphnetin,umbelliferone,apigenin,and genkwanin in the Daphne giraldii injection raw materials simultaneously.Methods The separation was performed on Luna C;(2)100 A(250 mm×4.6 mm,5μm)column,0.05%formic acid as mobile phase A,methanol as mobile phase B,gradient elution.Detection wavelength was set at 326 nm.The injection volume was 20μL,the column temperature was 35℃,the flow rate was 1.0 mL·min^(-1).Results The calibration curve was linear within the range of 3.0-300.0,3.0-300.0,2.0-200.0,and 2.0-200.0 mg·L^(-1)for daphnetin,umbelliferone,apigenin,and genkwanin,respectively,with the correlation r>0.9990.The recovery ranges were 99.2%,98.7%,98.1%,and 99.4%,respectively.The relative standard deviations(RSDs)were 1.2%,1.7%,1.6%,and 1.3%(n=6),respectively.Conclusion The established method is suitable for study the constents of daphnetin,umbelliferone,apigenin,and genkwanin in Daphne giraldii injection raw materials.
