摘 要:目的探究西洋参-石菖蒲对糖尿病大鼠海马线粒体形态结构及GLUT-4蛋白表达的影响。方法以链脲佐菌素构建糖尿病大鼠模型,分为糖尿病组(DM Group)、二甲双胍组(Metformin Group)、西洋参-石菖蒲组(X-S Group);STZ注射9 d后给予西洋参-石菖蒲药液每日1次,连续75 d;血糖仪检测FBG;Morris水迷宫评价各组大鼠的学习记忆能力,筛选糖尿病认知障碍大鼠和非认知障碍大鼠;透射电镜观察海马线粒体形态结构;免疫组化法观察海马GLUT-4蛋白表达。结果与糖尿病组比较,西洋参-石菖蒲组大鼠空腹血糖及学习记忆能力均无显著变化(P>0.05),认知障碍发生率降低;与DACD组比较,西洋参-石菖蒲组大鼠海马神经元及神经毡线粒体肿胀或固缩现象减轻,但海马神经元胞膜GLUT-4蛋白表达无显著变化。结论西洋参-石菖蒲可能通过海马线粒体的保护作用,降低糖尿病大鼠认知障碍发生率,而与促进海马GLUT-4蛋白膜转位无关。Objective To investigate the effects of Panax quinquefolium L.-Acorus tatarinowii on the morphological structure of hippocampus mitochondria and the expression of GLUT-4 protein in diabetic rats.Methods Construction of diabetic rat model with streptozotocin(STZ),It was divided into diabetes group(DM Group),metformin group(Metformin Group),Panax quinquefolium-Acorus calamus group(X-S Group);STZ administration of Panax quinquefolium-Acorus calamus once a day after 9 d of injection,75 d;continuous Blood glucose detector FBG;Evaluation of learning and memory ability of rats in Morris water maze,Screening of diabetic cognitive impairment(DACD)rats and non-cognitive impairment rats(DNCD);The morphology and structure of hippocampus mitochondria were observed by transmission electron microscope;Expression of GLUT-4 protein in hippocampus was observed by immunohistochemistry.Results Compared with diabetic group,There were no significant changes in fasting blood glucose and learning and memory abilities in in rats in Panax quinquefolium L.-Acorus tatarinowii group(P>0.05),the incidence of cognitive impairment decreased.Compared with DACD group,the swollen or pyknotic of mitochondria in neurons and neuropil in hippocampal of rats in Panax quinquefolium L.-Acorus tatarinowii group were alleviated,but there was no significant change in the expression of GLUT-4 protein in the cell membrane of neurons in hippocampus.Conclusions Panax quinquefolium L.-Acorus tatarinowii may reduce the incidence of cognitive impairment in diabetic rats through the protective effect of hippocampal mitochondria,but not related to the improvement of GLUT-4 protein membrane translocation in hippocampi.
