摘  要:目的通过网络药理学研究祖卡木颗粒成分、靶点、通路和生物学功能,初探其抗新型冠状病毒的作用机制。方法运用在线数据库获取祖卡木颗粒的主要活性成分及对应靶点,并利用CTD和DiSGeNET数据库获取新冠肺炎的疾病靶点。药物与疾病靶点去映射,用Cytoscape软件草药-成分-靶点多维网络,并用其ClueGO插件对映射的靶点进行基因功能富集和信号通路分析。结果筛选得到槲皮素、山柰酚、木犀草素、芦荟大黄素、甘草查尔酮A和异鼠李素等98个活性成分及其白介素、丝裂原活化蛋白激酶、趋化因子、γ-干扰素以及肿瘤坏死因子等222个靶点。疾病靶点有621个,药物与疾病靶点取交集得到88个潜在靶点。GO生物学功能富集得653个和KEGG通路富集得110条与COVID-19相关(P<0.005),涉及癌症和病毒相关的信号通路以及Toll样受体信号通路、细胞凋亡、IL-17信号通路和VEGF信号通路等炎症反应和免疫系统。结论从多维可视化网络角度分析来看,新疆祖卡木颗粒通过其多成分-多靶点来调控机体的病毒感染和免疫炎症反应。祖卡木颗粒针对新冠肺炎的病毒复制和细胞因子风暴具有显著的调节作用。本研究从一个方面阐明了以新疆祖卡木颗粒抗新冠病毒的作用机制,并为后期研究提供了理论依据。Objective The novel coronavirus was studied by network pharmacology studies on the composition,target,pathway and biological function of Zukamu granules.Methods The main active components and corresponding targets of Zukamu granules were obtained by online database,and The novel coronavirus pneumonia target was gained from CTD and DiSGeNET databases.Drugs and disease targets were mapped,and the multi-dimensional network of herbs,ingredients and targets was established by using the software of Cytoscape,Finally,we used its ClueGO plug-in to enrich the gene function and analyze the signal pathway of the intersection targets.Results We obtained 98 active components including Quercetin,Kaempferol,Luteolin,Aloe emodin,Liquorice chalcone A and Isorhamnetin,and 222 targets including Interleukin,Mitogen activated protein kinase,Chemokines,Interferon-γand Tumor necrosis factor.There were 621 disease targets,and 88 potential targets were received by intersection of drugs and disease targets.653 GO and 110 KEGG pathways were associated with COVID-19(P<0.005),involving cancer and virus related signaling pathways,as well as inflammatory response and immune system such as toll like receptor signaling pathway,apoptosis,IL-17 signaling pathway and VEGF signaling pathway.Conclusion From the perspective of multi-dimensional visualization network,Xinjiang Zukamu granules regulate virus infection and immune inflammatory response through its multi-component and multi-target.The novel coronavirus pneumonia has significant regulatory effects on viral replication and cytokine storm in new crown pneumonia.This study elucidated the mechanism of Xinjiang Zukamu Granule against new coronavirus from one aspect,and provided theoretical basis for later research.
