摘  要:目的研究香烟烟雾颗粒(DSP)对大鼠肠系膜动脉平滑肌内皮素受体的影响及作用机制。方法采用离体血管环技术观察DSP对内皮素受体介导的肠系膜动脉平滑肌收缩功能的影响,Western blot法检测内皮素受体以及MAPK和NF-κB信号通路相关蛋白的表达。结果DSP可以增强内皮素受体激动剂引起的血管收缩并使内皮素受体蛋白表达增加,ERK1/2和P38通路抑制剂可以抑制DSP诱导的血管收缩和降低内皮素受体的表达。结论DSP通过MAPK和NF-κB信号通路上调内皮素受体诱导大鼠肠系膜动脉平滑肌收缩。Objective To determine the effect of cigarette smoking particles on the vasoconstrictions induced by endothelin receptors in the mesenteric artery of rats.Methods In vitro aorta ring culture was used to observe how cigarette smoking particles affected vasoconstrictions induced by endothelin receptors and MAPK/NF-κB pathway related proteins expression by Western blot.Results Cigarette smoking particles enhanced mesenteric artery constriction induced by endothelin receptor agonists and increased the expression of endothelin receptors,while the inhibitors of ERK1/2 and P38 signaling pathway inhibited the vasoconstriction responses and decreased the protein expression of endothelin receptors.Conclusion Cigarette smoking particles can up-regulate endothelin receptor-mediated vaso contraction by activating MAPK and NF-κB signaling pathways in the mesenteric artery of rats.
