栀子花舒心胶囊、酸橙花舒畅胶囊和酸橙栀子双花抗脑衰胶囊对大鼠局灶性脑缺血/再灌注损伤的保护作用

摘　　要：目的探讨栀子花舒心胶囊、酸橙花舒畅胶囊和酸橙栀子双花抗脑衰胶囊对大鼠局灶性脑缺血/再灌注损伤的保护作用。方法将132只雄性SD大鼠随机分为11组:假手术组,模型对照组,脑心通胶囊组(2.01 g·kg^(-1)),酸橙花舒畅胶囊低、中、高(0.47、0.94、1.88 g·kg^(-1))剂量组,栀子花舒心胶囊低、中、高(0.63、1.26、2.52 g·kg^(-1))剂量组,酸橙栀子双花抗脑衰胶囊中、高(1.45、2.90 g·kg^(-1))剂量组。采用改良Zea Longa线栓法构建大鼠局灶性脑缺血/再灌注损伤模型,各给药组在造模手术前连续给药8 d,造模手术后进行神经功能缺陷评分,干湿称重法检测大鼠脑指数,TTC染色法检测大鼠脑梗死体积,试剂盒法检测大鼠血清SOD、MDA、谷氨酸、NO、Ca^(2+)的含量,酶联免疫法检测大鼠血清中炎症介质IL-6、IL-8、IL-1β、TNF-α的变化,HE染色和免疫组化观察海马皮质层区病理学改变。结果与模型组比较,各给药组能明显降低脑缺血/再灌注大鼠翻正反射恢复时间、降低脑组织含水量、减少脑梗死体积,提高SOD活性,降低MDA、谷氨酸、NO和Ca^(2+)含量,抑制激活小胶质细胞释放IL-6、IL-8、IL-1β、TNF-α炎症因子,改善病理形态学。结论栀子花舒心胶囊、酸橙花舒畅胶囊和酸橙栀子双花抗脑衰胶囊对局灶性脑缺血/再灌注损伤具有明显保护作用,其机制可能与抗炎及抗氧化有关。Objective To determine the effect of Gardenia flower Shuxin capsules,Lime flower Shuchang capsules and Gardenia lime flower Kangnaoshuai capsules on the anti-focal cerebral ischemia/reperfusion injury in rats.Methods Totally 132 male SD rats were randomly divided into 11 groups:a sham operation group,a model group,a Naoxintong capsule group(2.01 g·kg^(-1)),3(low,medium and high-dose)Lime flower Shuchang capsule groups(0.47,0.94,and 1.88 g·kg^(-1)),3(low,medium and high-dose)Gardenia flower Shuxin capsule groups(0.63,1.26,and 2.52 g·kg^(-1)),2(medium and high-dose)Gardenia lime flower Kangnaoshuai capsule groups(1.45,2.90 g·kg^(-1)).The focal cerebral ischemia/reperfusion model in rats with modified suture-occluded method of Zea Longa was established.Each group was ig administrated once daily for 8 days.The neurological function scores and brain indexes were evaluated,and the cerebral infarction area and various related enzymes(SOD,MDA,glutamate,NO,Ca^(2+),IL-6,IL-8,IL^(-1)β,and TNF-α)were determined.HE staining and immunohistochemistry were used to evaluate the cerebral tissue morphology.Results Compared with the model group,the medication groups obviously decreased the recovery time of righting reflex in rats with cerebral ischemia/reperfusion,alleviated the pathological damage of ischemia reperfusion place,reduced the water content of the brain tissue,reduced the cerebral infarction volume,increased SOD activity and reduced the content of MDA,glutamate,NO and Ca^(2+),and inhibited the activation of microglia releasing IL-6,IL-8,IL^(-1)βand TNF-αinflammatory factors.The pathomorphology was significantly improved.Conclusion Gardenia flower Shuxin capsules,Lime flower Shuchang capsules and Gardenia lime flower Kangnaoshuai capsules have obvious protective effect on focal cerebral ischemia/reperfusion injury,whose mechanism may be related to the anti-inflammation and antioxidation functions.
