摘 要:目的研究肾复舒颗粒对糖尿病肾病小鼠的保护作用及可能机制。方法C57BL/6J雄性小鼠40只,随机分为正常对照组、模型组、肾复舒颗粒组和阳性药物组,每组10只。采用腹腔注射链脲佐菌素制备糖尿病肾病模型。药物组给予2.08 g·kg^(-1)的肾复舒颗粒提取物混悬液(0.5%CMC-Na),阳性药物组给予缬沙坦溶液(40 mg·kg^(-1)),其他组给予相同体积的0.5%CMC-Na溶液。药物干预12周后,收集24 h尿液、血液以及肾脏组织。HE、PAS以及Masson’s染色评价肾脏组织病理变化,蛋白质印迹法检测NF-κB、fibronectin、p-NF-κB(S536)、p-IKKα/β(ser176)、IKKα/β、TGF-β、p-Smad2(Ser250)和collagen4A1蛋白的表达。结果与模型组比较,肾复舒颗粒显著降低血尿素氮、血肌酐和24 h尿微量白蛋白水平;降低肾小球面积、系膜基质以及胶原沉积;降低肾脏组织中IL-6和TNF-α水平及p-NF-κB(S536)、p-IKKα/β(ser176)、p-Smad2(Ser250)、fibronetin和collagen4A1蛋白的表达。结论肾复舒颗粒可延缓糖尿病肾病的进展,其作用机制可能与抑制炎症和纤维化有关。Objective To determine the protective effect of Shenfushu granules(SFSG)on diabetic nephropathy(DN)in mice and possible mechanism.Methods Forty C57BL/6J mice were randomized into four groups:a normal control group,a model group,an SFSG group,and a positive drug group(10 in each group).The DN model was established by intraperitoneal injection of streptozotocin.The SFSG group received SFSG suspendsion in 0.5%CMC-Na at 2.08 g·kg^(-1).The positive drug group received valsartan(40 mg·kg^(-1)),and other groups were given 0.5%CMC-Na.The treatment lasted 12 weeks.The 24 h urine,blood,and kidney tissue were collected.Hematoxylin-eosin,periodic acid-Schiff,and Masson’s staining were used to evaluate the histopathological changes.The expressions of NF-κB,fibronectin,p-NF-κB(S536),p-IKKα/β(ser176),IKKα/β,TGF-β,p-Smad2(Ser250),and collagen4A1 were detected by Western blot.Results Compared with the model group,SFSG significantly decreased the blood urea nitrogen,creatinine and 24-h albuminuria level,attenuated mesangial matrix and collagen deposition,and decreased the glomerular area,the levels of IL-6 and TNF-αin the kidney tissue,and the expressions of p-NF-κB(S536),p-IKKα/β(ser176),p-Smad2(Ser250),fibronetin,and collagen4A1.Conclusion SFSG delays the development of DN,whose mechanism of action may be associated with inhibition of inflammation and fibrosis.
