摘  要:目的:观察海龙黄精散对微波辐射诱导的少弱精子症小鼠精子活力及对肿瘤坏死因子-α(TNF-α)、白介素-6(IL-6)、白介素-12(IL-12)的影响。方法:将40只小鼠随机分为空白组、模型组及海龙黄精散小、中、大剂量组,每组8只。微波辐射造模同时,海龙黄精散小、中、大剂量组分别以0.0075、0.015、0.03 g海龙黄精散灌胃,干预18 d。HE染色观察小鼠睾丸组织病理学改变,比较各组小鼠精子活力、Johnson评分及睾丸组织TNF-α、IL-6、IL-12水平。结果:干预后,模型组小鼠精液前向运动精子(PR)、非前向运动精子(NP)、精子总活力低于空白组(均P<0.05),海龙黄精散小剂量组小鼠NP较模型组升高(P<0.05),海龙黄精散中、大剂量组小鼠PR、NP、精子总活力高于模型组(均P<0.05)。与空白组比较,模型组小鼠Johnson评分下降(P<0.05);与模型组比较,海龙黄精散小、中、大剂量组小鼠Johnson评分升高(均P<0.05)。与空白组比较,模型组小鼠睾丸组织TNF-α、IL-6、IL-12水平升高(均P<0.05);与模型组比较,海龙黄精散小、中、大剂量组小鼠TNF-α、IL-12水平降低(均P<0.05);与模型组比较,海龙黄精散中、大剂量组小鼠IL-6水平降低(均P<0.05)。结论:海龙黄精散可提高微波辐射诱导少弱精子症小鼠精子活力,其机制可能与海龙黄精散降低睾丸TNF-α、IL-6、IL-12水平有关。Objective:To observe the effects of Hailong Huangjing powder on sperm motility and TNF-α,IL-6,IL-12 in mice with oligoasthenospermia induced by microwave radiation.Methods:Forty mice were randomly divided into blank group,model group and Hailong Huangjing powder low-dose,medium-dose and high-dose groups,with eight mice in each group.At the same time of microwave radiation modeling,Hailong Huangjing powder low-dose,medium-dose and high-dose groups were given 0.0075,0.015,0.03 g of Hailong Huangjing powder,lasting 18 days.HE staining was used to observe the histopathological changes of testis,and sperm motility,Johnson score and the levels of TNF-α,IL-6 and IL-12 in testicular tissue of each group were compared.Results:After intervention,the sperm PR,NP and total sperm motility of model group were lower than those of blank group(all P<0.05),sperm NP in Hailong Huangjing powder low-dose group was higher than that in model group(P<0.05),sperm PR,NP and total sperm motility in Hailong Huangjing powder medium-dose and high-dose groups were higher than those in model group(all P<0.05).Compared with blank group,Johnson score of model group decreased(P<0.05).Compared with model group,Johnson score of Hailong Huangjing powder low-dose,medium-dose and high-dose groups increased(all P<0.05).Compared with blank group,the levels of TNF-α,IL-6 and IL-12 in testicular tissue of model group were increased(all P<0.05);compared with model group,the level of TNF-αand IL-12 in Hailong Huangjing powder low-dose,medium-dose and high-dose groups were decreased(all P<0.05).Compared with model group,the level of IL-6 in Hailong Huangjing powder medium-dose and high-dose groups decreased(all P<0.05).Conclusion:Hailong Huangjing powder can improve the sperm motility of mice with oligoasmatospermia induced by microwave radiation,and the mechanism may be related to the decrease of TNF-α,IL-6 and IL-12 levels in testis.
