复方苦参注射液和血必净注射液对大鼠急性肺损伤的作用对比研究

摘　　要：目的:探讨复方苦参注射液与血必净注射液对大鼠急性肺损伤的作用。方法:将Wistar大鼠随机分为正常组、模型组、阳性对照(地塞米松注射液)组、血必净注射液组及苦参高、中、低剂量组。除正常组外,其余各组均采用脂多糖(LPS)建立急性肺损伤大鼠模型,连续3 d。造模后立即给药,末次造模后15 h给予第4次给药。给药后1 h处死动物,取肺灌洗液和肺组织,采用酶联免疫吸附试验(ELISA)法检测肺灌洗液和组织匀浆中白介素-1β(IL-1β)、白介素-6(IL-6)、肿瘤坏死因子(TNF-α)和核因子(NF)-κB的含量,苏木精—伊红(HE)染色观察肺组织病理学变化。结果:与正常组比较,模型组肺组织炎性细胞浸润程度升高,阳性对照组、血必净注射液组、苦参高剂量组肺组织炎性细胞浸润程度低于模型组。与模型组比较,苦参高、中剂量组和血必净注射液组肺灌洗液和肺组织中IL-1β、IL-6、NF-κB和TNF-α水平降低(均P<0.05);与血必净注射组比较,苦参注射液中剂量组肺灌洗液和肺组织中NF-κB、TNF-α水平降低(P<0.05)。结论:复方苦参注射液和血必净注射液在治疗急性肺损伤方面均有一定的效果,能够有效抑制炎症反应,且前者的效果更为明显。Objective:To investigate the effects of compound Kushen injection and Xuebijing injection on acute lung injury in rats.Methods:Wistar rats were randomly divided into normal group,model group,positive control(dexamethasone injection)group,Xuebijing injection group and Kushen injection high,medium and low dose groups.Except for the normal group,the models of acute lung injury were established by lipopolysaccharide(LPS)inhalation for 3 d.The drug was administered immediately after modeling,and the fourth dose was given 15 h after the last modeling.The animals were executed 1 h after drug administration,with lung lavage fluid and tissues collected.The contents of interleukin-1β(IL-1β),interleukin-6(IL-6),tumor necrosis factor(TNF-α)and nuclear factor(NF)-κB in lung lavage fluid and tissues were measured by enzyme-linked immunosorbent assay(ELISA),and hematoxylin-eosin(HE)staining was used to observe the pathological changes of the lung tissues.Results:Compared with the normal group,the degree of inflammatory cell infiltration in the lung tissues was increased in the model group,and the degree of inflammatory cell infiltration in the lung tissues was lower in the positive control group,the Xuebijing injection group and the Kushen injection high-dose group than that in the model group.Compared with the model group,the levels of IL-1β,IL-6,NF-κB and TNF-αin lung lavage fluid and lung tissues were decreased in the Kushen injection high and medium dose groups and the Xuebijing injection group(P<0.05).Compared with the Xuebijing injection group,the levels of NF-κB and TNF-αin lung lavage fluid and lung tissues were decreased in the Kushen injection medium dose group(P<0.05).Conclusion:Both compound Kushen injection and Xuebijing injection are effective in the treatment of acute lung injury and can effectively suppress inflammatory response,and the former has better effects.
