摘 要:目的:建立702糖浆中何首乌、鸡血藤、钩藤的薄层色谱鉴别及何首乌中有效成分2,3,5,4’-四羟基二苯乙烯-2-O-β-D葡萄糖苷(简称二苯乙烯苷)的含量测定方法,为702糖浆的质量控制提供依据。方法:采用薄层色谱法(TLC)定性鉴别702糖浆中的何首乌、鸡血藤、钩藤等成分,采用高效液相色谱法(HPLC)测定二苯乙烯苷的含量。结果:何首乌、鸡血藤、钩藤等的TLC斑点清晰、阴性对照无干扰,具有较强的特异性;二苯乙烯苷在1.0146~25.0190μg/mL范围内与峰面积呈现良好线性关系(r=0.9999),平均加样回收率为99.6%,RSD为1.58%;各批样本间二苯乙烯苷含量比较,差异无统计学意义(P>0.05)。结论:该方法可以定性鉴别702糖浆并测定其有效成分的含量,且操作简便,重复性好,稳定性高,可用于702糖浆的质量控制。Objective:To establish a TLC identification of Polygonum multiflorum,Caulis Spatholobi,Uncaria in 702 syrup and the content determination method of 2,3,5,4’-tetrahydroxystilbene-2-O-β-D glucoside(referred to as stilbene glycoside)in Polygonum multiflorum to provide an reference for the quality control of 702 syrup.Methods:The components of Polygonum multiflorum,Caulis Spatholobi,Uncaria in 702 syrup were qualitatively identified by thin layer chromatography(TLC).The content of stilbene glycoside was determined by high performance liquid chromatography(HPLC).Results:The TLC spots of Polygonum multiflorum,Caulis Spatholobi,Uncaria,etc,were clear,the negative control had no interference,and had strong specificity.Stilbene glycoside showed a good linear relationship between the peak area and the range of 1.0146-25.0190μg/mL(r=0.9999),the average recovery rate was 99.6%,and the RSD was 1.58%.There was no significant difference in the content of stilbene glycoside among different batches of samples(P>0.05).Conclusion:This method can qualitatively identify 702 syrup and determine its effective components,and it has the advantages of simple operation,good repeatability and high stability,and can be used for the quality control of 702 syrup.
