摘  要:目的观察“退热六法”对不同月龄脂多糖致热幼兔的退热效果及TLR4/NF-κB信号通路表达的影响。方法选取1、2、3月龄新西兰幼兔随机分为正常组、模型组、推拿组,每组8只。推拿组和模型组通过耳缘静脉注射0.5 μg·kg^(-1)LPS建立发热模型,1 h内肛温升高0.6℃为造模成功,推拿组于造模后1 h进行手法干预,干预方法为退热六法,即开天门、推坎宫、揉太阳、揉耳后高骨、清天河水、推脊,连续监测6h肛温变化,观察不同月龄幼兔的退热效果,选取效果最优的2月龄幼兔,观察血浆和下丘脑TNF-α、IL-1β蛋白含量、下丘脑中TLR4 NF-κB p65蛋白含量、下丘脑中TLR4、NF-κB、TNF-α、IL-1βmRNA水平。结果“退热六法”干预后,1月龄肛温最低,2月龄次之,3月龄显著高于1月龄(P<0.05);1月龄降低肛温最大幅度显著大于2月龄和3月龄(P<0.05);2月龄发热抑制时程显著长于1月龄和3月龄(P<0.05)。推拿组血浆和下丘脑中TNF-α、IL-1β含量显著低于模型组(P<0.05);推拿组TLR4、细胞核NF-κB p65表达显著低于模型组(P<0.05),推拿组下丘脑TLR4、NF-κB、TNF-α、IL-1βmRNA显著低于模型组(P<0.05)。结论“退热六法”降温作用明确,1月龄降温幅度最大,2月龄次之;2月龄降温最稳定,3月龄次之。“退热六法”可能是通过抑制TLR4/NF-κB途径起到抗炎解热作用。Objective To observe the antipyretic effect of the six methods for antipyretic on the lipopolysaccharide-induced fever in young rabbits of different months and its influence on the expression of the TLR4/NF-κB signaling pathway.Methods One-,two-,and three-month-old New Zealand young rabbits were randomly divided into a normal group,a model group,and a massage group,with 8 rabbits in each group.In the massage group and model group,a fever model was established by injecting 0.5 μg·kg^(-1) LPS into the ear marginal vein.The model was successfully established when the anal temperature increased by 0.6 ℃ within one hour.In the massage group,the six methods for antipyretic were performed one hour after modeling,namely Kai Tianmen,Tui Kangong,Rou Taiyang,Rou Erhou Gaogu,Qing Tianheshui and Tui Ji.The change of anal temperature was continuously monitored for 6 hours to observe the antipyretic effect in the young rabbits of different months.The two-month-old rabbits with the best effect were selected to observe the protein contents of TNF-α and IL-1β in plasma and hypothalamus,the protein contents of TLR4 and NF-κB p65 in hypothalamus,and the mRNA levels of TLR4,NF-κB,TNF-α,IL-1β in hypothalamus.Results After the intervention,the anal temperature was the lowest in the one-month rabbits,followed by the two-month rabbits,while the temperature in the three-month-old rabbits was significantly higher than that in those of the one-month rabbits (P<0.05).The maximum temperature reduction in the one-month rabbits was significantly greater than that in the two-month rabbits or three-month rabbits (P<0.05).The duration of fever inhibition of the two-month rabbits was significantly longer than those of the one-month and three-month rabbits (P<0.05).The contents of TNF-α and IL-1β in plasma and hypothalamus of the massage group were significantly lower than those of the model group (P<0.05).The expression levels of TLR4 and nuclear NF-κB p65 in the massage group were significantly lower than those in the model group (P<
