摘  要:目的初步探讨抗Siglec-9抗体Fab段在小鼠巨噬细胞上对LPS诱导的炎症反应的作用及其机制。方法将小鼠巨噬细胞分为对照组、LPS组、LPS+Ab组、Ab组,采用实时荧光定量PCR技术检测TNF-α、IFN-β、IL-1、IL-6等炎性细胞因子的mRNA水平;采用ELISA检测炎性细胞因子的蛋白水平;通过Western blot初步探讨抗Siglec-9抗体Fab段影响炎性细胞因子表达量的作用机制。结果与LPS组相比,抗Siglec-9抗体Fab段抑制炎性细胞因子的mRNA和蛋白水平,降低TLR4信号通路磷酸化水平。结论抗Siglec-9抗体Fab段通过阻断TLR4信号传导通路抑制炎性细胞因子的m RNA和蛋白水平,在感染性疾病中具有潜在的应用价值。The study was designed to investigate the effect of human anti-Siglec-9 antibody Fab fragment on LPS induced inflammatory response of mouse macrophages.Mouse macrophages were divided into control group,LPS group,LPS+Ab group,Ab group.Quantitative real-time PCR and enzyme-linked immunosorbent assay were used to evaluate the mRNA and protein expression of inflammatory cytokines TNF-α,IFN-β,IL-1,and IL-6.While Western blot was used to measure the phosphorylation level of TLR4 signaling pathways.Data showed that the anti-Siglec-9 antibody Fab fragment considerably reduced the mRNA and protein expression levels of inflammatory cytokines,and could significantly suppressed the phosphorylation of the TLR4 signaling pathway proteins.In conclusion,the anti-Siglec-9 antibody Fab fragment can inhibit the mRNA and protein expression levels of inflammatory cytokines by downregulating the TLR4 signaling pathway,thus the anti-Siglec-9 antibody Fab fragment could be a potential anti-inflammatory drug in the treatment of inflammatory disease.
